Derivation of multipotent nestin+/CD271−/STRO-1− mesenchymal-like precursors from human embryonic stem cells in chemically defined conditions

Abstract: The successful establishment of stem cell-based therapies requires multipotent, immunocompatible stem cells, highly efficient strategies for direct differentiation, and most importantly, optimal culture conditions for large-scale expansion of such cell populations. Other than adult tissues, human embryonic stem cells (hESCs) represent another infinitely expansible source for mesenchymal stem cell (MSC) derivation. Here, we reproducibly derived a population of Nestin(+)/CD271(-)/STRO-1(-) mesenchymal-like precu… Show more

“…We have not included nestin, Sox2, or Oct4, in the analysis, since these are intracellular proteins but not surface markers. However, we noticed that nestin(+)/CD271(−)/Stro‐1(−) MSCs derived from human ESCs were reported to differentiate into representative derivatives of all three embryonic germ layers . Therefore, compared with CD271 and Stro‐1, nestin positivity may represent a more primitive phenotype of MSCs.…”

“…Increased expression of Klf 4 and c myc, ESC specific transcription fac tors, has also been found . The expres sion of undifferentiated ESC markers in MSCs proba bly explains their higher differentiation potential (Wu et al, 2013). Enhanced expression of these markers in spheroids compared to monolayer cultures suggests that they are primitive subpopulations with multipo tent potential (Bogdanova Jatniece et al, 2014;Guo et al, 2014).…”

“…This strategy is supported by step-wise protocols to achieve MSC differentiation and then chondrogenesis. To date, several reports have described the derivation of MSCs from hESCs for subsequent chondrogenic differentiation and cartilage tissue engineering (99102). The implanted hESC-derived mesenchymal cells gave rise to homogeneous, well-differentiated tissues exclusively of mesenchymal origin and without teratoma formation (103).…”
