Der Sonnenblumen‐Trypsin‐Inhibitor 1 (SFTI‐1) in der Chemie und Biologie

Die Kernspinresonanz (NMR) hat sich zu einer universellen Methode für biochemische und biomedizinische Studien, einschließlich Metabolomik, Proteomik und Magnetresonanztomographie (MRT), entwickelt. Durch die Vervielfachung des NMR‐Signals ausgewählter Moleküle hat die Kernspin‐Hyperpolarisation das Anwendungsspektrum von NMR und MRT bedeutsam erweitert (z. B. hyperpolarisierte Festkörper‐NMR und metabolische Bildgebung in vivo). Parawasserstoff (pH2) erlaubt es, Moleküle schnell und kostengünstig zu hyperpolarisieren. In diesem Gebiet wurden in den letzten zehn Jahren umfangreiche Fortschritte erzielt, einschließlich der Synthese neuer Tracer, Katalysatoren und Methoden für die Polarisationsübertragung. Das Portfolio an hyperpolarisierten Molekülen umfasst nunmehr Aminosäuren, die für viele Anwendungen von großem Interesse sind. In diesem Beitrag beleuchten wir die aktuelle Literatur zur Hyperpolarisation von Aminosäuren und Peptiden.

Ein bizyklisches Peptid wurde mit einem Diarylethen modifiziert, um lichtschaltbare Inhibitoren der Serinprotease Bos taurus Trypsin 1 (T1) zu erzeugen. Ausgehend von dem Sonnenblumen‐Trypsin‐Inhibitor‐1 (SFTI‐1) als Prototyp, erhielten wir lichtschaltbare Inhibitoren von T1 mit Ki im niedrigen nanomolaren Bereich, deren Aktivität durch Bestrahlung mit Licht um das 20‐Fache moduliert werden konnte. Die inhibitorische Potenz sowie die Resistenz gegen proteolytischen Abbau wurden systematisch an einer Reihe von 17 SFTI‐1‐Analoga untersucht. Die Isomerisierung des Photoschalters beeinflusst das Netzwerk aus Wasserstoffbrücken, das zur Stabilisierung der Struktur der Inhibitoren dient, und möglicherweise auch die Bindungsdynamik zwischen Enzym und Inhibitor. Die Machbarkeit einer zeitlichen und räumlichen Manipulation der Enzymaktivität wurde durch kontrollierten Verdau eines Hydrogels auf Gelatinebasis und eines antimikrobiellen Peptids BP100‐RW bewiesen. Die angewendeten Designprinzipien für Diarylethen‐Photoschalter stellen eine Grundlage für die Entwicklung anderer Serinprotease‐Inhibitoren dar.

Nuclear magnetic resonance (NMR) has become a universal method for biochemical and biomedical studies, including metabolomics, proteomics, and magnetic resonance imaging (MRI). By increasing the signal of selected molecules, the hyperpolarization of nuclear spin has expanded the reach of NMR and MRI even further (e.g. hyperpolarized solid‐state NMR and metabolic imaging in vivo). Parahydrogen (pH2) offers a fast and cost‐efficient way to achieve hyperpolarization, and the last decade has seen extensive advances, including the synthesis of new tracers, catalysts, and transfer methods. The portfolio of hyperpolarized molecules now includes amino acids, which are of great interest for many applications. Here, we provide an overview of the current literature and developments in the hyperpolarization of amino acids and peptides.
