Purpose Some drug products contain phthalates as excipients, and in vitro studies have demonstrated that phthalates interfere with cellular mechanisms involved in colorectal cancer development. We therefore examined the association between cumulative phthalate exposure from drug products and risk of colorectal adenocarcinomas. Methods We used the Danish Cancer Registry to identify all patients with incident colorectal adenocarcinoma from 2008 to 2015 (n = 25 814). Each cancer case was matched to ten population controls. Linking information from Danish registers, we quantified cumulative phthalate exposure to the ortho‐phthalates diethyl phthalate (DEP) and dibutyl phthalate (DBP) as well as enteric phthalate polymers from orally administered drugs. The association between cumulative phthalate exposure and colorectal cancer was estimated using conditional logistic regression. Results Cumulative exposure to ortho‐phthalates exceeding 500 mg was associated with lower odds of colorectal cancer diagnosis (ORadj = 0.89; 95% CI, 0.81‐0.96). Similar associations were observed for all DEP exposure exceeding 500 mg. Subgroup analysis excluding NSAID users, demonstrated that ortho‐phthalate exposure was positively associated with colorectal cancer (ORadj = 1.26; 95% CI, 1.05‐1.51). Conclusion We found an apparent overall protective effect of cumulative phthalate exposure from drug excipients for colorectal adenocarcinoma. Omitting NSAID users reversed the signal and suggested a slightly increased risk associated with high cumulative ortho‐phthalate exposure.

Resveratrol is a type of polyphenol that is abundantly present in knotweed and grapes, and has been confirmed to have tumor-inhibitory properties. However, the effect of resveratrol on tumor cells under chronic stress conditions remains unclear. The aim of the present study was to demonstrate that resveratrol exerts a significant inhibitory effect on the growth and proliferation of tumor cells under chronic stress in a dose-dependent manner. Furthermore, resveratrol was able to induce apoptosis of cancer cells under chronic stress conditions. Moreover, resveratrol was found to inhibit tumor cells under chronic stress by decreasing the expression of the β 2 -adrenergic receptor (ADRB-2); in addition, the protein expression of hypoxia-inducible factor (HIF)-1α was suppressed by resveratrol in a dose-dependent manner. Thus, these data suggest that resveratrol inhibits the growth of cancer cells under chronic stress by regulating the ADRB-2-HIF-1α axis. In summary, the present study may provide a new basis supporting the use of resveratrol for the prevention and treatment of pancreatic cancer.

Background: Evidence from observational studies suggests a protective role for physical activity (PA) against colorectal cancer (CRC) risk. However, it has yet to be established a causal relationship. We conducted a two-sample Mendelian randomisation (MR) study to examine causality between physical activity and CRC risk. Methods:We used common genetic variants associated with self-reported and accelerometerbased physical activity as instrumental variables (IVs) in this MR study. The IVs were derived from the largest available genome-wide association study (GWAS) of physical activity, namely UK Biobank. We analysed the effect of the IVs for physical activity in a large CRC GWAS that included 31 197 cases and 61 770 controls. We applied inverse variance weighted (IVW) method as the main analysis method.Results: Our results demonstrate a protective effect between accelerometer-based physical activity and CRC risk (the outlier-adjusted ORIVW was 0.92 per one standard deviation (SD) increase of accelerometer-base physical activity [95% CI: 0.87-0.98, P: 0.01]). The effect between self-reported physical activity and CRC risk was not statistically significant but was also supportive of an inverse association (the outlier-adjusted ORIVW was 0.61 per 1 SD increase of moderate-to-vigorous physical activity [95%CI: 0.36-1.06, P: 0.08]). Conclusions:The findings of this large MR study show for the first time that objectively measured physical activity is causally implicated in reducing CRC risk. The limitations of the study are that it is based on only two genetic instruments and that it has limited power, despite the study size. Nonetheless, at a population level, these findings provide strong reinforcing evidence to support public health policy measures that encourage exercise, even in obese individuals.
