Depletion of calcium stores regulates calcium influx and signal transmission in rod photoreceptors

Abstract: Tonic synapses are specialized for sustained calcium entry and transmitter release, allowing them to operate in a graded fashion over a wide dynamic range. We identified a novel plasma membrane calcium entry mechanism that extends the range of rod photoreceptor signalling into light-adapted conditions. The mechanism, which shares molecular and physiological characteristics with store-operated calcium entry (SOCE), is required to maintain baseline

“…After sequential image acquisition of cells' fluorescence at 495/440 nm or 340/380 nm, the backgrounds were subtracted using commercial software (Metafluor; Universal Imaging, West Chester, PA and NIS Elements, Melville, NY). Calibration of [Ca 2ϩ ]i and pHi was carried out as described previously (63)(64)(65)(66). Intracellular pH was calibrated from 495-to-440 ratios using the nigericin/high extracellular K ϩ method (10 M nigericin/90 mM KCl).…”

“…Nevertheless, the typical phenotypes of CNG channel deficiency are impaired opsin trafficking and opsin mislocalization (8,20), reflecting impaired ER function/protein folding, and ER stress. It is worth mentioning that there are other Ca 2ϩ channels in the inner segment membrane, including transient receptor potential channels, and these channels could also play a role in the cytosolic Ca 2ϩ regulation (42,43). entry is probably decreased) to compensate for the reduced cytosolic Ca 2ϩ level, which could potentially lead to a reduction of ER Ca 2ϩ level.…”

“…In mouse retina, TRPC1 was expressed in rods and cones and rod store-operated Ca 2+ entry was unaffected in Trpc1-null mice, indicating that TRPC1 does not regulate light-evoked responses at the mouse rod output synapse (Molnar et al 2012). However, downregulation of TRPC1 by RNAi did suppress storeoperated Ca 2+ entry in rods (Szikra et al 2008). The reasons for these discrepancies are unknown but could stem from inherent differences between acute (RNAi in vitro) and chronic (knockout in vivo) gene ablation.…”
