Dependence on calcium concentration and stoichiometry of the calcium pump in human red cells

Abstract: SUMMARY1. In resealed human red cells loaded with Ca-EGTA buffer solutions it was found that the intracellular free Ca2+ concentration for half saturation of the Ca transport system (which pumps Ca out of the cell) is equal to or smaller than 4 x 10-6 M and thus closely agrees with the dissociation constant of the Ca+ Mg activated membrane ATPase.2. The maximal rate of Ca transport from resealed cells to medium was found to be 0 148 + 0 009 #zmole/ml. cells.min at 280 C.3. The rate of Ca transport was unaffect… Show more

“…However, according to their results, ϳ1.5 mol of Ca 2ϩ are occluded per mole of enzyme. This value is in contrast with the proposed stoichiometry of 1 Ca 2ϩ transported per ATP hydrolyzed by the PMCA (20,21,22). In our hands 4 (not shown, but see Ref.…”

“…Calcium transport was conducted by the membrane filtration technique (2) in a total volume of 0.5 ml containing 50 mM Tris-Mes (pH 7.5), 50 mM KCI, 5 Estimation of Ionized Calcium Concentration. The ionized calcium concentration was calculated using an EGTA buffer and successive approximation analysis as described by Schatzmann (25). The stability constant for the Ca24-EGTA complex used for calculations was 10"°' and the protonization values a, and a2 were 109.38 and 10877, respectively, giving an association constant at pH 7.5 of 107.83 (20).…”

“…For determination of kinetic parameters, Ca-EGTA buffers were used. Concentrations of free CaZf at pH 6.8 were calculated with an apparent stability constant for Ca-EGTA of 105.94 according to Schatzmann [37].…”
