The study describes procedures that were used to develop a highly sensitive enzyme-linked immunosorbent assay (ELISA) for the quantitation of a trichothecene mycotoxin, 3-acetyldeoxynivalenol (3-AcDON), in barley. Polyclonal antibodies were produced in rabbits immunized with a conjugate of 3-AcDON and human serum albumin linked by an ester linkage (hemisuccinate bridge). High anti-3-AcDON titers were obtained after multiple immunizations. However, only a negligible degree of inhibition was obtained with 3-AcDON in a competitive ELISA when the coating conjugate contained the same ester linkage group (hemiglutarate bridge) as the immunogen. The use of a conjugate containing a heterologous ether linkage (O-methylcarboxyl bridge) compared to that of the immunogen yielded an inhibition curve for 3-AcDON that was highly sensitive (IC50 = 0.21 ng/ml) with essentially no interference from the bridging group. This conjugate was synthesized using iodoacetate and 1,1′-carbonyldiimidazole chemistries. The assay showed low cross-reactivity with other trichothecenes including several analogs of deoxynivalenol (DON) with the exception of acetylated DON. The ELISA developed on the basis of this new conjugate was able to detect low concentrations of 3-AcDON (16 ppb) in spiked barley without any cleanup treatments.

Aim: To determine whether assessing the penetration of solutions with different concentrations of ethanol (alcohol percentage test: APT) on fungal surfaces is effective in characterization of hydrophobicity on fungal surfaces. Methods and Results: APT and contact angle (CA) measurements were conducted on nine hydrophobic and two hydrophilic fungal strains from the phyla of Ascomycota, Basidiomycota and Zygomycota. There was a strong positive correlation (R2 = 0·95) between the APT and CA measurements from eight of the nine hydrophobic stains (four pathogenic and mycotoxigenic Fusarium taxa, one melanosporaceous biotrophic taxon, Alternaria sp, Penicillium aurantiogriseum and Cladosporium cladosporioides). Hydrophilic control strains, Mortierella hyalina and Laccaria laccata, had CAs <90° and no measurable degree of hydrophobicity using the APT method. Conclusions: The APT method was effective in measuring the degree of hydrophobicity and can be conducted on different zones of fungal growth. Significance and Impact of the Study: Characterization of fungal surface hydrophobicity is important for understanding of its particular role and function in fungal morphogenesis and pathogenesis. APT is a simple method that can be utilized for fungal hydrophobicity measurements when CA cannot be measured because of obscured view from aerial mycelia growth.

Problem statement: Identification of Quantitative Trait Loci (QTLs) in durable resistant genotype is important for marker-assisted breeding. The study was conducted to identify QTLs linked to FHB resistance in 'Tokai 66', a Japanese genotype. Approach: A cross was made between Tokai 66 and Jagalene and the single seed descend was used to advance the population. The F 2:4 and F 2:5 populations were evaluated by artificially inoculating disease in a mist-irrigated nursery in 2006 and 2007. Disease incidence, severity, FDK and DON content were evaluated in the 128 RILs. Map Manager QTX was used to prepare the linkage map and QTL Cartographer 2.0 was used to identify QTLs for FHB responses from the field data respectively. Results: Four QTLs for FHB responses were detected, of which one QTL each for severity and FDK were stable across two years and were located at the 5B and 3BSc, respectively. Both QTLs conferred resistance to FHB. Two unstable QTLs were detected at the 5B and 3D for FDK and DON content, respectively using the field environment.The 5B QTL for FDK contributed to susceptibility, whereas the 3D QTL for DON content contributed to resistance. Conclusion: The 5B QTL associated with resistance to severity and 3B QTL associated with resistance to FDK could be utilized by winter wheat breeding programs selection to enhance FHB resistance.
