Anaerobic oxidation of methane (AOM) reduces methane emissions from marine ecosystems, but we know little about AOM in freshwater reservoirs. Lake Baikal is the world’s only freshwater reservoir where gas hydrates (GH) are found. Despite that AOM has been demonstrated in deep sediments of Lake Baikal did not reveal methane-oxidizing archaea ANME1, 2, or 3, which are responsible for AOM in marine sediments. A search for representatives Candidatus Methylomirabilis oxyfera (M. oxyfera)-like bacteria (NC10) and Candidatus Methanoperedens nitroreducens (M. nitroreducens)-like archaea (ANME-2d) has been carried out in the different types of Lake Baikal methane enriched sediments. We used different molecular biology methods including high-throughput sequencing and PCR analysis, using 16S rRNA genes as well as different functional genes of AOM (mcrA and pmoA). We found diverse M. oxyfera-like bacteria and M. nitroreducens-like archaea in various geological structures in Lake Baikal (methane seep and mud volcano), which were different from the composition of the discharged fluid. We also considered possible electron acceptors for this process in the sediments of Lake Baikal.

Abstract. Imamsyah A, Arthana IW, Astarini IA. 2020. The influence of physicochemical environment on the distribution and abundance of mangrove gastropods in Ngurah Rai Forest Park Bali, Indonesia. Biodiversitas 21: 3178-3188. Ngurah Rai Forest Park is the widest mangrove ecosystem in Bali that close to the business center and tourism area. The strategic location of the Ngurah Rai Forest Park is estimated to produce anthropogenic waste that can disrupt the gastropod population and stability of the mangrove ecosystems. This study aims to analyze the mangrove density, distribution, and abundance of gastropods based on the quality of the biophysical environment. Mangrove data collection was carried out using a plot transect of 10 m x 10 m, 5 m x 5 m, and 1 m x 1 m. Meanwhile, samples gastropods were collected on a plot transect of 0.5 m x 0.5 m in plot transect of 5 m x 5 m. A total of 11 gastropods species and 6 mangrove species were recorded in this study. The dominant gastropods species found were Assiminea brevicula (28 ind/m2). Species mangrove of Sonneratia alba and Rhizophora apiculata found were in all study sites with tree density values of 1000 ind/ha. Subsequently, the ecological index calculation results show that the diversity index (H’) (2.89-3.2), evenness index (E) (0.87-0.93), and dominance index (C) (0.12-0.17). Based on the Principal Component Analysis (PCA) found was Assiminea brevicula spread on sand and silt substrates and adaptive to the temperature condition, salinity, pH, and dissolved oxygen. Furthermore, other analysis results showed that Cerithidea cingulata, Cerithidea quadrata, Littoraria articulata, and Littoraria scabra were found on clay substrate with high C-organic content. In conclusion, gastropods are evenly distributed and no species dominate the ecosystems.

Background The discovery of complete ammonia oxidizer (comammox) was groundbreaking. Comammox can use ammonia as the sole nitrogen source and turn it to nitrate. Moreover, genomic data indicated that comammox contained genes which can metabolize urea and nitrite. However, the feasibility of enriching comammox with urea and nitrite in long term has not been proved. This study enriched comammox’s culture by using nitrite in reactor SA and urea in reactor SB. Results The nitrification rate of reactor SB (1.29 mg N·g -1 biofilm · d -1 ) was higher than that in reactor SA (0.6 mg N · g -1 biofilm · d -1 ) at the 390 th day. Comammox outnumbered ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) in both reactor SA (9.04 × 10 9 copies / g biofilm) and reactor SB (5.34×10 10 copies/ g biofilm). In reactor SA, comammox’s amoA accounted for 92% of the total amoA, which was higher than that in reactor SB (85%). However, the percentage of comammox (4%) in total bacteria was much lower than reactor SB (14%). The results of metagenomic sequencing showed that all the pathways of nitrogen cycle including nitrification, nitrogen fixation, denitrification, assimilation nitrate reduction, and dissimilation nitrate reduction can be detected in both reactor SA and reactor SB except the anammox pathway. The genes related to nitrite oxidation and nitrate reduction in reactor SA (TPM = 5099; TPM = 3329) was higher than that of in reactor SB (TPM = 4071; TPM = 2984), presumably due to the demand of turning nitrite to nitrate and turning nitrate to ammonia. While genes related to ammonia oxidation and urea metabolism in reactor SB (TPM = 3915; TPM = 3638) was higher than that in reactor SA (TPM = 2708; TPM = 3002). Conclusion Nitrite and urea can regulate the enrichment culture of comammox by converting its metabolic pathway. Using nitrite as sole nitrogen source can improve the proportion comammox’s amoA in total amoA while using urea as the sole nitrogen source may increase comammox’s proportion in total bacteria. These results can accelerate the enrichment of comammox and facilitate the promotion of comammox’s engineering operation.
