Dengue subgenomic RNA binds TRIM25 to inhibit interferon expression for epidemiological fitness

Abstract: The global spread of dengue virus (DENV) infections has increased viral genetic diversity, some of which appears associated with greater epidemic potential. The mechanisms governing viral fitness in epidemiological settings, however, remain poorly defined. We identified a determinant of fitness in a foreign dominant (PR-2B) DENV serotype 2 (DENV-2) clade, which emerged during the 1994 epidemic in Puerto Rico and replaced an endemic (PR-1) DENV-2 clade. The PR-2B DENV-2 produced increased levels of subgenomic f… Show more

“…Three days after dsRNA injection, 24-h-starved mosquitoes were offered a blood meal comprised of 40% washed erythrocytes from specific-pathogen-free pig's blood (PWG Genetics), 5% 5 mM ATP (Thermo Fisher Scientific), 5% human serum (Sigma), and a 50% volume of RPMI containing virus diluted to 10 7 PFU/ml. Blood titers were validated in a plaque assay as described previously (69). The infectious blood meal was maintained at 37°C using a hemotek feeder system (Discovery Workshops) covered by a stretched pig's intestine.…”

“…Mosquitoes were allowed to feed for 2 h. Engorged females were selected and maintained with access to sugar in the insectary. Six days post-oral infection, individual mosquitoes were homogenized in 500 l of RPMI, filtered through a 0.22-m filter (Sartorius) and determined the titer of by plaque assay (69).…”

“…In mammalian cells, sfRNA is essential for flavivirus-induced cytopathicity and pathogenicity (25). Importantly, sfRNA acts as an antagonist of both interferon-and retinoic acid-inducible gene-I-like receptor-dependent innate immune responses (31,32). Accordingly, WNV mutants deficient in sfRNA production are attenuated in mice (25,29).…”

“…This sub genomic RNA seems to be responsible for regulating the cellular cytosolic receptor RIG-I that recognizes viral RNA. This signaling pathway would be similar to the one adopted by Dengue virus [17].…”
