Dendritic Cells and Their Role in Immunotherapy

Abstract: Despite significant advances in the field of cancer immunotherapy, the majority of patients still do not benefit from treatment and must rely on traditional therapies. Dendritic cells have long been a focus of cancer immunotherapy due to their role in inducing protective adaptive immunity, but cancer vaccines have shown limited efficacy in the past. With the advent of immune checkpoint blockade and the ability to identify patient-specific neoantigens, new vaccines, and combinatorial therapies are being evaluat… Show more

“…cDC1 is primarily defined by the transcription factors BATF3 and IRF8. Type 1 cDCs are also defined by cell-surface expression of CD8α and CD103, with CD8α + cDC1s considered to be lymphoid organ-resident cDC1s, and CD103 + cDC1s considered to be tissue-resident or migratory cDC1s ( 5 ). cDC2 is frequently defined by the transcription factor IRF4 and surface expression of CD11b, CD1c, and SIRPα ( 5 ).…”

“…Whereas cDCs and pDCs are derived from common DC progenitors, these subsets possess considerably different characteristics and functions ( 5 ). As mentioned above, antigen presentation is the main role of cDCs.…”

“…Sometimes, the DCs are fused to BC cells in vaccine production [ 76 , 77 ]. Moreover, adjuvants such as IL-12, IFN-γ, lipopolysaccharides (LPS), and other cocktails are used to induce a mature DC phenotype [ 78 , 79 , 80 , 81 , 82 ]. These pulsed and primed DCs are then inoculated back into the patient or animal model as a vaccine.…”
