Dendritic Cells and Myeloid Derived Suppressor Cells Fully Responsive to Stimulation via Toll-Like Receptor 4 Are Rapidly Induced from Bone-Marrow Cells by Granulocyte-Macrophage Colony-Stimulating Factor

Abstract: Dendritic cells (DCs) are commonly generated from bone marrow (BM) progenitor cells with granulocyte-macrophage colony-stimulating factor (GM-CSF) alone or in combination with interleukin 4 (IL-4). These cells are often harvested post day 5, when they acquire maturation markers and can stimulate T cells. Apart from DCs, myeloid derived suppressor cells (MDSCs) are also found within these cultures. However, little is known about the functional characteristics of DCs and MDSCs before day 5. Herein, using a murin… Show more

“…Monocytes are crucial players in the innate immune system and as such, rely on PRRs to detect pathogens and initiate host defence responses. Amongst these are TLRs, which play a vital part in host defence response and activation, direct microbiocidal activity, and activate inflammatory pathways [ 109 , 110 ]. TLRs on the surface of cells recognise microbial membrane components, whilst intracellular TLRs recognise bacterial and viral nucleic acids.…”

“…PAMPs recognized by surface TLRs induce more mature DCs (mDCs) to stimulate specific anticancer immune responses. These well-studied PAMPs include lipopolysaccharide (LPS) and GM-CSF (detected by TLR4) [94] , [95] , [96] , bacterial lipoproteins and lipoteichoic acids (detected by TLR2), flagellin (TLR5 agonist) [97] and Pam3CSK4 (TLR2 agonist) [98] . SRA/CD204 is a PRR that acts as a negative immune regulator via downregulation of TLR4.…”

“…In vitro, studies have proved that the DCs are an important component during osteoclast development where they have used CD11c + CD11b − DC subsets for the development of osteoclasts (Li et al, 2008). These subsets were derived from primary bone marrow cultures in the presence of GM‐CSF and IL‐4 stimulation (Kong et al, 2020). The mature osteoclasts formed were actively functional and efficiently induced bone resorption.…”
