Dendritic cells and cytokines in human inflammatory and autoimmune diseases

Abstract: Dendritic cells (DCs) produce cytokines and are susceptible to cytokine-mediated activation. Thus, interaction of resting immature DCs with TLR ligands, for example nucleic acids, or with microbes leads to a cascade of pro-inflammatory cytokines and skewing of T cell responses. Conversely, several cytokines are able to trigger DC activation (maturation) via autocrine, for example TNF and plasmacytoid DCs, and paracrine, for example type I IFN and myeloid DCs, pathways. By controlling DC activation, cytokines r… Show more

“…9, No. 3; induce their effects on the immune system by direct cell-cell contact or by secretion of multiple cytokines including IL-1, IL-6, IL-12 (to which IL-23 belongs), tumor necrosis factor-ɑ and interfron-ɑ/β (Blanco et al, 2008). High serum and cervical levels have been observed for some of the cytokines (such as IL-6 and IL-23) in women with URSA (Galazios et al, 2011;Hattori et al, 2007).…”

“…To identify this specific role of W-PG we assayed multiple cytokines and chemokines since the cytokines released from DCs are known to enable not only paracrine activations to lymphocytes but also autocrine activations to themselves. 31,32) A significant difference was observed in the secretion of certain cytokines between the stimulation with MIX and MIXϩW-PG (Table 1). iDCs express a unique repertoire of inflammatory chemokine receptors such as CCR2, CCR3, and CCR5.…”

“…Langerhans cells were characterised as permissive cells for DENV in vivo replication (Wu et al 2000). Indeed, DCs have a crucial role in initiating host defence mechanisms; through antigen presentation and cytokine production, they can define the fate of the immunological response (Blanco et al 2008). Both DENV and YF vaccine viruses are known to infect DCs and monocytes in vitro (Wu et al 2000, Barba-Spaeth et al 2005, Reis et al 2007).…”
