Dendritic Cell-Specific Delivery of Flt3L by Coronavirus Vectors Secures Induction of Therapeutic Antitumor Immunity

Abstract: Efficacy of antitumor vaccination depends to a large extent on antigen targeting to dendritic cells (DCs). Here, we assessed antitumor immunity induced by attenuated coronavirus vectors which exclusively target DCs in vivo and express either lymphocyte- or DC-activating cytokines in combination with a GFP-tagged model antigen. Tracking of in vivo transduced DCs revealed that vectors encoding for Fms-like tyrosine kinase 3 ligand (Flt3L) exhibited a higher capacity to induce DC maturation compared to vectors de… Show more

“…Overall, analysis of these data indicated that substantial changes in intranodal T cell migration occurred when >70% of FRCs were lost, i.e., at DT doses of !2 ng/g. To assess how FRCs affect DC-mediated activation of antiviral CD8 + T cells, we resorted to a viral vector system that facilitates exclusive in vivo targeting of DCs [40,41]. Propagation-deficient coronavirus particles were injected subcutaneously into FRCdepleted mice, and the activation of antiviral CD8 + T cells was assessed in draining LNs.…”

“…As far as defects in innate immunity are concerned, only rather obscure models such as IFNAR1-deficient mice and TLR7-deficient mice were studied. Polymorphisms in those genes were associated with severe or unusual COVID-19 in humans, but it is unclear whether those models are truly relevant [ 70 , 96 , 150 , 151 ]. Inborn errors of acquired immunity are also associated with severe COVID-19 [ 152 ].…”

“…That same group also demonstrated that vectors encoding Flt3L, exhibited a higher capacity to induce DC maturation compared to vectors delivering IL-2 or IL-15. The former more efficiently induced tumor-specific CTLs with expanded epitope repertoire, resulting in therapeutic tumor immunity ( 146 ).…”
