Dendritic Cell Maturation Induced by Muramyl Dipeptide (MDP) Derivatives: Monoacylated MDP Confers TLR2/TLR4 Activation

Abstract: 6-O-acyl-muramyldipeptides (MDP) with various lengths of fatty acid chains were examined for their dendritic cell (DC) maturation activity expressed through TLRs. Judging from anti-TLR mAb/inhibitor-blocking analysis, MDP derivatives with a single octanoyl or stearoyl fatty acid chain were found to activate TLR2 and TLR4 on human DCs, although intact and diacylated MDP expressed no ability to activate TLRs. Human DC activation profiles by the monoacylated MDP were essentially similar to those by Calmette-Guér… Show more

“…For instance Natural Killer (NK) cells, which belong to Group 1 ILCs and attend in the gut homeostasis by responding to commensally enteric bacteria through the innate immune system and cytolytic activity, express high levels of NOD2 [106], and MDP treatment of these cells leads directly to cell activation through the NOD2-mediated pathway [107]. In addition, MDP can also be sensed by TLR2 (described below) [108,109] which is expressed on RORγt + ILCs [110]. Therefore, MDP might have an indirectly influence on ILCs through modulation of other cellular components of the innate immunity.…”

“…Beyond the ability to activate NOD2-mediated signaling pathways, MDP can interact and signal through TLR2 [108,109] . TLR2 recognizes MDP and activates NEMO-NF-κB pathway through two independently upstream signaling pathways [120].…”

“…2A). Since interaction between NOD2 and specific TLR pathways has been reported as a mechanism of cooperation in the innate immune response, which might lead to the synergistic activation of host cells [24][25][26], the effect of the combined addition of both Staphylococcus aureus PGN and MDP was assessed. As shown in Fig.…”
