Demonstration of primary cilia and acetylated α-tubulin in fish endothelial, epithelial and fibroblast cell lines

Abstract: Primary cilia (PC) were demonstrated for the first time in fish endothelial, epithelial and fibroblast cell lines through immunofluorescence staining with the monoclonal antibody, 6-11B-1, against acetylated α-tubulin. The study was carried out with eight recently developed cell lines from the walleye, Sander vitreus (Mitchill). These were three fibroblast-like cell lines, WE-cfin11f, WE-skin11f and WE-liver3 from, respectively, the caudal fin, skin and liver, and three epithelial-like cell lines, WE-cfin11e, … Show more

“…As part of studying senescence (Vo, Mikhaeil, Lee, Pham, & Bols, ) and primary cilia in fish cell lines (Vo & Bols, ), we developed a walleye skin fibroblastoid cell line, WE‐skin11f, but did not characterize the cells further. WE‐skin11f is the first cell line from normal walleye skin and so further characterization is warranted.…”

“…Transmembrane proteins, polycystin-1 and polycystin-2, form a complex at the ciliary membrane to mediate calcium influx. Geerts et al, 2011;Egorova et al, 2012) and fluorescent confocal microscopy (Van der Heiden et al, 2006;Geerts et al, 2011;Russellpuleri et al, 2017;Nauli et al, 2008;Antal et al, 2017;Vo and Bols, 2016;Iomini et al, 2004;Ki et al, 2020;Dummer et al, 2018;Goetz et al, 2014). Transmission electron microscopy can provide the detailed ultrastructure information of the cilium and scanning electron microscopy can provide information regarding cell surface morphology, cilia density and the contact of the cilia with the cell surface.…”

“…Tubulin and microtubules (MTs) are the largest cytoskeletal components ( 16 ), and posttranslational modifications of tubulin are found in all cells with MTs ( 17 , 18 ). For example, acetyl-α-tubulin regulates MT stabilization and cell morphology ( 19 , 20 ). In addition, the loss of acetyl-α-tubulin is associated with TGF-β-induced epithelial-mesenchymal transition and sulfur mustard-induced chronic airway remodeling ( 21 , 22 ).…”
