Demonstration of Equine Herpes Virus 1 (EHV‐1) in Histological Sections and Tissue Cultures by the Peroxidase‐Antiperoxidase (PAP) Technique

Abstract: Summary The demonstration of EHV‐1 antigen in formalin‐fixed and paraffin‐embedded liver tissue sections of aborted foetuses is described. The peroxidase‐antiperoxidase (PAP) technique is easy to perform and compared to the immunofluorescent technique seems to be more useful, especially if slightly autolysed material is being investigated. “…In accordance with the findings of other authors (Whitwell, 1982;Gimeno et al, 1987;Edington et al, 1991), antigen detection was found to be slightly more sensitive than VI in this study, as in two cases virus could be detected only by IH. The primary site of virus replication is the fetal lung (Westerfield and Dimock, 1946).…”

“…Because they cannot be considered diagnostic for the virus infection, diagnosis should be supported by a specific laboratory test (Schultheiss et al, 1993). The VI technique is time consuming and expensive; additionally, false-negative results may be caused by post-mortem changes (Gimeno et al, 1987;Galosi et al, 2001). IH is relative cheap, fast and seems to be more sensitive than the VI method.…”

“…The diagnosis in Argentina of EHV‐1 infection associated with abortion in mares has traditionally been based on the presence of histopatological changes and VI in cell culture (Gimeno et al., 1987). In this study, it was demonstrated that the PCR method described is rapid, sensitive and specific.…”

“…Routine clinicopathology may reveal leucopenia (Paradis 1996). A virus-specific diagnosis is made through virus culture, specific antibody titre .increaae (seroconversion) and detection of antigen through immunofluorescence, immunocytochemistry (Merkel and Zimmer 1981;Mumford 1984;Allen and Bryans 1986;Gimeno et at. 1987;van Maanen et at.…”
