Demonstrating approaches to chemically modify the surface of Ag nanoparticles in order to influence their cytotoxicity and biodistribution after single dose acute intravenous administration

Abstract: With the advance in material science and the need to diversify market applications, silver nanoparticles (AgNPs) are modified by different surface coatings. However, how these surface modifications influence the effects of AgNPs on human health is still largely unknown. We have evaluated the uptake, toxicity and pharmacokinetics of AgNPs coated with citrate, polyethylene glycol, polyvinyl pyrolidone and branched polyethyleneimine (Citrate AgNPs, PEG AgNPs, PVP AgNPs and BPEI AgNPs, respectively). Our results d… Show more

“…Citrate-coated AgNPs have a higher affinity to proteins than PVP-coated AgNPs, suggesting they could absorb more proteins on their surface (Ahlberg et al, 2014). However, the amount of protein (serum albumin) absorbed on AgNPs was similar in citrate- and PVP-coated AgNPs (Pang et al, 2016). In addition, protein corona composition was similar for citrate- and PVP-coated AgNPs after AgNP incubation in cell culture medium supplemented with fetal bovine serum (Shannahan et al, 2013).…”

“…Some predictive models of nanoparticles uptake by cells and apoptosis were developed and a strategy for modeling was applied for various nanoparticles or modifications of their surfaces. (Epa et al, 2012;Pang et al, 2016;Zhang et al, 2012). A model to describe toxic potencies of 17…”

“…Compared with 20-and 100-nm PVPcoated AgNPs and 110-nm citrate-coated AgNPs, 20-nm citrate-coated AgNPs induced the highest levels of oxidative stress and necrosis (10). Furthermore, particle coating (polyethylene glycol vs. branched polyethyleneimine) was also observed to modify toxicity of AgNPs in the Hep-a1c1c7 mouse liver cell line (11).…”
