Delivering Enhanced Testosterone Replacement Therapy through Nanochannels

Abstract: Primary or secondary hypogonadism results in a range of signs and symptoms that compromise quality of life and requires life-long testosterone replacement therapy. In this study, an implantable nanochannel system is investigated as an alternative delivery strategy for the long-term sustained and constant release of testosterone. In vitro release tests are performed using a dissolution set up, with testosterone and testosterone:2-hydroxypropyl-β-cyclodextrin (TES:HPCD) 1:1 and 1:2 molar ratio complexes release … Show more

“…For example, in particular nanochannelar membranes (e.g. silicon) have been explored for drug delivery applications with great results [38][39][40]. However, the substrate material is generally silicon and immunogenicity studies are as a result performed on nanochanneled silicon surfaces [41].…”

“…Intracellular signaling and stimuli, as well as cell-to-cell and cell-to-matrix physiological transport, can be effectively mimicked by nanofluidic systems 3 , 4 . Such systems are also currently utilized in multiple fields, including desalination 5 , 6 , cell and drug delivery 7 9 , analyte sorting 10 , and filtration 11 , among others. Unique transport behaviors can be attained when diffusion occurs under nanoscale spatial confinement, especially when the size of the fluidic systems approaches the size of fluid molecules.…”

“…This control is relevant to a broad spectrum of biomedical applications, including drug delivery (Desai et al 1999; Ferrati et al 2013, In press; Celia et al 2014), molecular sieving (Fu et al 2007), cell sorting (Gossett et al 2010), and cell transplantation (Desai et al 1998; Sabek et al 2013). In drug delivery, the ability to fine tune release profiles for specific therapeutic agents is highly beneficial in personalized treatments requiring dose titration or synchronization with the biological clock, the fundamental principle in chronotherapy (Smolensky and Peppas 2007; Youan 2010).…”

“…This platform has been specifically designed for hormone replacement, organ transplantation, and cancer prevention, among others (Fine et al 2010; Ferrati et al 2013, In press; Sih et al 2013). We hypothesized that drug release through our membrane could be modulated by using a low-voltage electrical potentials.…”
