Delineation of HER2 Gene Status in Breast Carcinoma by Silver in Situ Hybridization is Reproducible among Laboratories and Pathologists

Abstract: An automated enzyme metallographic silver in situ hybridization method (SISH) has been reported to successfully determine human epidermal growth factor receptor 2 (HER2) gene amplification. We evaluated the staining and interpretative reproducibility of the HER2 SISH assay at five laboratories and compared SISH results with other in situ hybridization (ISH) methods. The HER2 gene status of 89 breast carcinomas was analyzed in parallel using manual dual-color fluorescence ISH , manual chromogenic ISH , and brig… Show more

“…These results are consistent with previous studies reporting on concordance rates for HER2 silver enhanced in situ hybridization vs. fluorescence in situ hybridization of 96% (k ¼ 0.75, 95% CI) and 99%. 13,27 Additionally, we assessed the EGFR gene status with a semiquantitative polymerase chain reactionbased assay and compared the results with silver enhanced in situ hybridization. Semiquantitative polymerase chain reaction-based assay results concurred excellent with silver enhanced in situ hybridization findings (Po0.001) and showed high association with EGFR silver enhanced in situ hybridization.…”

“…Bu yöntem kantitatif bir yöntem olup değerlendiriciler arasındaki uyum yüksektir. Ayrıca SİSH ASCO/CAP kılavuzunun önerdiği floresan in situ hibridizasyon (FISH) ile %95 uyumlu olma kriterine uygundur (17).…”

“…Until now, some studies have been performed to compare FISH and SISH methods with a correlation ratio of 89-98% (Dietel et al, 2007;Carbone et al, 2008;Papouchado et al, 2010;Ying et al, 2010;Park et al, 2011;Lee et al, 2012;Tvrdík et al, 2012). A correlation ratio of 92.3% was found in this study between FISH and SISH.…”

“…As a result, HER2 amplification in neoadjuvant therapy is directly related to the response to trastuzumab therapy. Baseline HER2/neu status identification with protein and/or genomic status is performed with immunohistochemistry and in situ hybridization (ISH), respectively (Dietel et al, 2007;Carbone et al, 2008;Francis et al, 2009;Bilous et al, 2012 to the fluorescence in situ hybridization (FISH) method, which is a generally accepted and commonly used in situ hybridization technique, and to compare the results of each method. HER2 and CEP17 status can be evaluated simultaneously with the newly developed "Dual-Color Probe".…”
