Delineation by use of specific monoclonal antibodies of the T-cell receptor and major histocompatibility complex interaction sites on the superantigen toxic shock syndrome toxin 1

Abstract: Murine monoclonal antibodies (MAbs) specific for toxic shock syndrome toxin 1 (TSST-1), a bacterial superantigen, showed the ability either to detect TSST-1 bound to histocompatibility locus antigen (HLA)-DR molecules or to inhibit TSST-1 binding to HLA-DR. A MAb capable of detecting DR-bound TSST-1 could also inhibit the toxin-induced activation of a T-cell receptor V␤15-expressing murine T-cell hybridoma. Alternatively, MAbs with specificity for the HLA-DR association site could present TSST-1 in vitro, stim… Show more

“…The mouse hybridoma producing the anti-idiotypic monoclonal IgG1 anti(anti-K13) (clone 5868C [10]) was kindly provided by Dr K. Stein, NIH, Bethesda, MD. The control clone (IgG1 anti-toxic shock syndrome toxin-anti-TSST, clone 49A3.7 [12]), was kindly provided by Dr A. Tarkowski, Göteborg.…”

“…In this study, three anti-SEB mAbs (B327, 2B33, and B344; reference 23 ), two anti-SEA mAbs (A116.3 and A108.6; Kappler, J., and P. Marrack, unpublished data), and two anti– TSST-1 mAbs (TS327H1.1 and 477L; reference 24 ) were used. The anti–TSST-1 mAbs were a gift of M. Matsumura (NeXstar Pharmaceuticals, Lakewood, CO).…”

“…The concentration of immunoglobulins was determined by the radial immunodiffusion method [7]. These MoAbs discriminate various binding sites of the TSST-1 molecule [8]. MAbA5 is directed at an MHC class II binding domain of human monocytes, and neutralizes TSST-1induced lethality in D-galactosamine-sensitized BALB/c mice [9].…”

“…In the next three experiments antibodies 93E1.2 and 769U1.1 were used alone or in combination and administered after bacterial injection. They were chosen because of their property to inhibit TSST-1-mediated T cell stimulation and to inhibit TSST-1 binding to HLA-DR, respectively [8]. The antibodies were administered 48 h after bacterial injection and then every second or every third day until the mice were killed.…”
