Deletion of the Prdm3 Gene Causes a Neuronal Differentiation Deficiency in P19 Cells

Abstract: PRDM (PRDI-BF1 (positive regulatory domain I-binding factor 1) and RIZ1 (retinoblastoma protein-interacting zinc finger gene 1) homologous domain-containing) transcription factors are a group of proteins that have a significant impact on organ development. In our study, we assessed the role of Prdm3 in neurogenesis and the mechanisms regulating its expression. We found that Prdm3 mRNA expression was induced during neurogenesis and that Prdm3 gene knockout caused premature neuronal differentiation of the P19 ce… Show more

“…However, the neuronal-specific role of PRDM3 remains to be addressed in mammals, and PRDM3 could be implicated in the onset of neurogenesis. Recently, our study showed that P19 cells with Prdm3 gene knock-out displayed earlier maturation of neurons along with the rapid proliferation of non-neuronal cells [102]. These findings strongly showed the significant role of PRDM3 in the formation of the mammalian nervous system in vivo.…”

“…Gene knock-out leads to precocious neuronal differentiation in the P19 cells (direct mechanism unknown, increased expression of MAP2 and β-III TUBULIN). [102] Hamlet (Drosophila melanogaster PRDM3 and PRDM16 homolog) removes notch-dependent fate signature during neuronal-class diversification via direct chromatin-modification.…”

“…Like Neuro2a cells, ACSF2 had a Golgi specific localization in mouse P19 teratocarcinoma cells, as indicated by colocalization with Golgin 97 (Fig. 5A-C); both Neuro2a cells and P19 cells have been studied as models for neurogenesis [24]. In contrast to Neuro2a and P19 cells, however, mouse MA10 (testicular Leydig) cells (Fig.…”

“…The ATTO-550 ribonucleoprotein CRISPR cas9 (RNP) complex was combined with Lipofectamine® CRISPRMAX™ (Thermo Fisher Scientific, Inc.) by incubation at 4°C for 30 min, and the mixture was subsequently used for transfection. A highly efficient procedure ( 30 ) was followed precisely for the transfection experiments.…”
