Deletion of the carboxyl‐terminal residue disrupts the amino‐terminal folding, self‐association, and thermal stability of an amphipathic antimicrobial peptide

Abstract: Understanding the complex relationship between amino acid sequence and protein behaviors, such as folding and self-association, is a major goal of protein research. In the present work, we examined the effects of deleting a C-terminal residue on the intrinsic properties of an amphapathic α-helix of mastoparan-B (MP-B), an antimicrobial peptide with the sequence LKLKSIVSWAKKVL-NH2. We used circular dichroism and nuclear magnetic resonance to demonstrate that the peptide MP-B([1-13]) displayed significant unwind… Show more

“…HQNO can also induce S. aureus to adopt the SCV phenotype (Hoffman et al, 2006 ) and this may also affect AMP activity. The effect of high and low temperatures on storage or preparation of AMP, as well as activity, has received consideration in a number of studies (Wei et al, 2007 ; Zhang et al, 2011 ; Ji et al, 2014 ; Lee et al, 2014 ; Jiao et al, 2019 ). For example, many AMP demonstrate stability at temperatures below 100°C.…”

“…However, the application of α-helical AMPs has been hindered by some serious obstacles, such as systemic toxicities and high manufacturing costs. To address these issues, short AMPs derivatives with cell-selective toxicity have been developed based on the amino acid composition, charge, and hydrophobicity of natural AMPs7891011.…”

“…These results are consistent with the reported diverse roles of the C-and Ntermini of membrane-active peptides. [19] Based on the outcome of the glycine scan, we computationally generated 20 3 = 8000 variations of Decoralin residues 1, 4, and 11 by allowing each of the 20 standard amino acids in these three positions. We chose this permutation strategy because we wanted to modulate peptide activity through the smallest possible sequence modification with ah igh chance of retaining membrane activity.…”
