Deletion of LCE3C and LCE3B genes is associated with psoriasis in a northern Chinese population

Abstract: The findings indicate that the LCE3C_LCE3B-del is an important risk factor in the pathogenesis of psoriasis and that the LCE3C_LCE3B-del does not show an epistatic effect with the HLA-Cw6 allele on susceptibility to psoriasis in the northern Chinese population.

“…In this study, a population attributable risk of 23% was found, indicating that the deletion explains a sizable proportion of the genetic contribution. The association with LCE3C_LCE3B-del has been replicated in several cohorts with different ethnic backgrounds (Huffmeier et al, 2010a;Li et al, 2011;Riveira-Munoz et al, 2011;Xu et al, 2011). In addition, in an independent GWAS of a large Chinese cohort, SNPs in strong linkage disequilibrium with LCE3C_LCE3B-del were identified as risk factors for psoriasis (Zhang et al, 2009).…”

“…Another study showed that no significant epistatic effect was observed between LCE3C_LCE3B-del and PSORS1 in the 53 C. Shen et al / Journal of Genetics and Genomics 42 (2015) 49e56 Tunisian population (Chiraz et al, 2014). In two studies of the Chinese population, there was no evidence of any interaction between the LCE3C_LCE3B-del locus and HLA-Cw6 Xu et al, 2011). In Hüffmeier's study , interaction between LCE3C_LCE3B-del and PSORS1 in the German population was marginal and non-significant after correcting for testing of multiple interactions by Bonferroni correction, and no evidence was observed for interaction of LCE3C_LCE3B-del with the copy number variation (CNV) of b-defensins (DEFB) or with a variant of the interleukin 12B (IL12B) gene.…”

“…Thirty-nine studies were identified by electronic and manual searches, and 10 were selected for a full-text review based on the title and abstract details (de Cid et al, 2009;Coto et al, 2010;Huffmeier et al, 2010aHuffmeier et al, , 2010bDocampo et al, 2011;Li et al, 2011;Riveira-Munoz et al, 2011;Xu et al, 2011;Ammar et al, 2012;Bergboer et al, 2012). Three of these 10 studies were excluded because they contained duplicated data (de Cid et al, 2009;Huffmeier et al, 2010b;Bergboer et al, 2012).…”

“…The LCE3C_LCE3B-del polymorphism has been reported to be associated with psoriasis by some, but not all, researchers, possibly due to small sample sizes, low statistical power, and/or clinical heterogeneity (Coto et al, 2010;Huffmeier et al, 2010aHuffmeier et al, , 2010bDocampo et al, 2011;Li et al, 2011;RiveiraMunoz et al, 2011;Xu et al, 2011;Ammar et al, 2012). Therefore, to overcome the limitations of individual studies, resolve inconsistencies, and reduce the likelihood that random errors are responsible for false-positive or false-negative associations, we performed a meta-analysis Nath et al, 2005;Lee et al, 2006).…”
