Deletion of Dictyostelium discoideum Sir2A impairs cell proliferation and inhibits autophagy

Abstract: Sirtuins are a family of deacetylases (Class III histone deacetylases) with evolutionarily conserved functions in cellular metabolism and chromatin regulation. Out of the seven human Sirtuins, the function of Sirt2 is the least understood. The purpose of the present study was to investigate the role of Sir2A, a homolog of human Sirt2 in Dictyostelium discoideum (Dd), a lower eukaryote. We created both overexpressing and deletion strains of Ddsir2A to analyse its functions. We observed sir2A mRNA expression thr… Show more

“…It contains a wider variety of histone variants that are more similar to vertebrates than other simple eukaryotic model organisms 44 46 . The major PTMs on histones are also observed in this organism, including ADPr 45 49 . Importantly, Dictyostelium also contains single copy histone genes that are amenable to genetic manipulation, opening up the possibility to perform gene replacement and site-specific mutation strategies to assess the functional significance of histone PTMs 45 , 50 , 51 .…”

“…Although the ability to phosphorylate S-T sites may be context dependent, we predict this strategy will be applicable to other ADPr sites in histones and other targets for Ser-ADPr. The conservation of the major histone PTMs in Dictyostelium 45 49 , together with the ability to generate site-specific mutations in histones, makes this an attractive system to study these possibilities.…”

“…Antisense RNA probe (DIG labeled; Roche Diagnostics, India) was synthesized by in vitro transcription from T3 promoter (a run off template generated by digestion with Sal I) and sense probe with T7 RNA polymerase (template generated by digestion with Bam HI). The probes were further used for hybridization in multicellular structures developed according to the protocol of Lohia et al ().…”
