Delayed Hypersensitivity-Type Granuloma Formation and Dermal Reaction Induced and Elicited by a Soluble Factor Isolated From Schistosoma Mansoni Eggs

Abstract: A soluble material has been isolated from schistosome eggs which in minute quantities without the addition of adjuvant induces sensitization to a delayed hypersensitivity type of granuloma forming around intact schistosome eggs. This material is secreted by intact eggs and is found in high concentration in the fluid released during the hatching process. When adsorbed to bentonite particles this substance elicits hypersensitivity type granuloma formation in specifically sensitized animals. The granuloma sensiti… Show more

“…Soluble extract from Toxoplasma gondii tachyzoites (STAg) and extract from Schistosoma mansoni eggs (SEA) were prepared as described (21,22). Recombinant proteins and Abs were purchased from Sigma-Aldrich, Cell Signaling, and BD Pharmingen except anti-IL-4 (11B.11), which was obtained from the National Institutes of Health Biological Resources Branch repository, and anti-ZAP70 Ab, which was kindly provided by Dr. Larry Samelson (National Cancer Institute, National Institutes of Health, Bethesda, MD) (23).…”

“…Sterile SEA was prepared according to standard procedures [46]. In brief, livers from chronically infected NMRI mice were homogenized and digested overnight with 1 mg/mL collagenase VIII (Sigma-Aldrich Chemie GmbH, Taufkirchen, Germany) and 312 U/mL DNAse (Sigma).…”

“…Parasite antigens, mitogens, antibodies and recombinant cytokines were added to the appropriate wells. Soluble egg antigen (SEA) and soluble adult worm preparation (SWAP) were used at a final concentration of 20 µg/mL and 50 µg/mL, respectively (Boros & Warren 1970, Pearce et al 1992. Phorbol myristate acetate (50 ng/mL) plus ionomycin (1 µg/mL) were used as mitogens.…”
