BACKGROUND: Fungicides are often applied to pears before they are kept in storage facilities. The scientific application of pesticides can reduce unnecessary exposure, which in turn could benefit both humans and the environment. RESULTS: We investigated dissipation behavior and residue distribution, and conducted risk assessments for prochloraz, pyraclostrobin, and tebuconazole in pears stored under different conditions using ultra-performance liquid chromatography (UPLC). The recoveries of the three fungicides ranged from 76.5% to 114.5%, and the coefficients of variation were 1.0%-8.5%. The half-life (t 1/2 ) ranges for degradation of the three fungicides in Dangshan Su pear peel were 8.8-13.9 days after storage at 25°C and 99.0-346.6 days after storage at 2°C. Among the three fungicides, tebuconazole had the lowest residue concentration in pear pulp (maximum of 0.226 mg·kg −1 ) and the longest half-life (≥ 231.0 days). Accordingly, among these fungicides, tebuconazole is the most suitable for the preservation of Dangshan Su pears during storage. Finally, we analyzed samples of six pear varieties from markets in China and found that the residue concentrations of the three fungicides in pear pulp and fruit met Chinese standards.CONCLUSION: The results provide a scientific basis for rationalizing the use of prochloraz, pyraclostrobin, and tebuconazole, and improving the safety of pears for eating.

The dissipation of field-applied difenoconazole, imidacloprid, pyraclostrobin and spinosad on Clementine mandarins (Citrus clementina Hort. ex Tan.) under controlled conditions throughout the citrus production chain was assessed. At harvest, 42 days after application, the dissipation of these pesticides were 80, 92, and 48% for difenoconazole, imidacloprid, pyraclostrobin, respectively, and spinosad was below the level of detectability. At day 28 after application, spinosad was no longer detected. The model equations that best describe the dissipation curves of these pesticides on Clementine mandarins showed different patterns. Their half-life on Clementine, calculated by the best-fitted experimental data, were 19.2 day (1st-order model) for difenoconazole, 4.1 day (Root Factor (RF) 1st-order model) for imidacloprid, 39.8 day (2nd-order model) for pyraclostrobin and 5.8 day (1st-order model) for spinosad. These results are the first record of pyraclostrobin persistence on mandarins, showing a longer half-life in this matrix than those reported for any other fruit. The treated fruit were harvested and submitted to the usual postharvest treatments: first, a hypochlorite drenching was performed; as a second step, imazalil and wax were applied, and then the mandarins were stored at 4 °C. After 32 days, cold storage caused no significant effects on the residue levels of the four pesticides compared with those determined on freshly harvested mandarins. All residues were below their Codex and European Union (EU) maximum residue limit (MRL) for mandarin since the spray application day.

Ortho-phenylphenol (OPP) is a fungicide contained in agro-industrial effluents produced by fruit-packaging plants. Within the frame of developing bio-strategies to detoxify these effluents, an OPP-degrading Sphingomonas haloaromaticamans strain was isolated. Proteins/genes with a putative catabolic role and bacterium adaptation mechanisms during OPP degradation were identified via genomic and proteomic analysis. Transcription analysis of all putative catabolic genes established their role in the metabolism of OPP. The formation of key transformation products was verified by chromatographic analysis. Genomic analysis identified two orthologous operons encoding the ortho-cleavage of benzoic acid (BA) (ben/cat). The second ben/cat operon was located in a 92-kb scaffold along with (i) an operon (opp) comprising genes for the transformation of OPP to BA and 2-hydroxypenta-2,4-dienoate (and genes for its transformation) and (ii) an incomplete biphenyl catabolic operon (bph). Proteomics identified 13 up-regulated catabolic proteins when S. haloaromaticamans was growing on OPP and/or BA. Transcription analysis verified the key role of the catabolic operons located in the 92-kb scaffold, and flanked by transposases, on the transformation of OPP by S. haloaromaticamans. A flavin-dependent monoxygenase (OppA1), one of the most up-regulated proteins in the OPP-growing cells, was isolated via heterologous expression and its catabolic activity was verified in vitro.
