Using stereological methods, especially the optical disector for unbiased estimation of nuclear number, our recent study demonstrated that long-term (6 or 12 months) vasectomy in the rhesus monkey had no significant effects on spermatogenesis (Peng et al. Reproduction 2002, 124, 847-856). This study aimed to determine the scenario in the rabbit using the same morphometric methodology. Three groups of normal male Japanese white rabbits (aged 4-5 months) were subjected to unilateral vasectomy; 10 days, 6 months and 12 months later both testes and epididymides were removed. Testicular and epididymal methacrylate-embedded sections were obtained for stereology.Vasectomy-induced damage to spermatogenesis was observed, primarily sloughing of spermatogenic cells with a greater reduction in the number of advanced (adluminal) cells. The damage was most severe at 10 days, occurring in all the testes on the vasectomized side and involving sloughing of even type A spermatogonia, the number of which returned to normal at 6 and 12 months. Damage was less severe at 6 and 12 months, being found in half of the testes of the vasectomy side, in which the total numbers of later germ cell types were 24.0-59.1% (spermatocytes) and 0.3-11.6% (spermatids) of control at 6 months, and 20.1-22.1% (spermatocytes) and 0.4-12.0% (spermatids) of control at 12 months. By contrast, Sertoli cell number per testis was unchanged following vasectomy in any group.Epididymis on the vasectomy side, especially at 10 days and 6 months, appeared larger than on the contralateral side, but this difference was not statistically significant, and no sperm granuloma was seen in the epididymis.

Bu çalışmada erişkin sıçanlarda vazektomi sonrası seminifer tübül epitelindeki yapısal değişiklikleri mikroskobik olarak incelemek ve nitel ve nicel olarak değerlendirmek amaçlanmıştır. Gereç ve Yöntemler: Toplam 48 adet 8 haftalık erkek Wistar albino sıçan kullanıldı. Çalışma deney grupları ve kontrol grupları içerdi (her grup için, n=8). Deney gruplarında tek taraflı vazektomi uygulanırken, kontrol gruplarında yalnızca abdominal kesi yapıldı. Deney ve kontrol gruplarındaki sıçanların sol testisleri vazektomiden 1, 12 ve 24 hafta sonra perfüzyonla alınarak Bouin fiksatifine kondu. Rutin histolojik takip işleminden sonra her gruptan ikişer testis Masson'un trikrom boyası ve orseinle boyanarak ışık mikroskobunda nitel olarak değerlendirildi. Altışar testis histomorfometrik inceleme için 1/8 oranında seri kesilerek PAS+hematoksilenle boyandı. Stereolojik inceleme için Cavalieri ve optik parçalama+optik disektör sondaları kullanıldı. Bulgular: Kontrol gruplarına kıyasla 1 ve 12 haftalık vazektomi gruplarında ortalama spermatogonyum, Sertoli ve miyoid hücre yoğunluğunun arttığı, spermatid sayısının ise azaldığı saptandı. Yirmi dört haftalık vazektomi grubunda ise seminifer tübülü oluşturan hücrelerin azaldığı görüldü. Nitel değerlendirmede vazektomi sonrası süreye paralel olarak ara maddedeki kolajen lif miktarında artış ve 1 haftalık deney grubunda seminifer tübül lümeninde dev hücre oluşumu gözlendi. Tartışma ve Sonuç: Vazektomi sonrasında 12. haftaya kadar seminifer tübül adlüminal ve bazal kompartımanlarında iki farklı mekanizmanın çalıştığı görülmüştür. İlkin, bazal kompartımanda gerek parakrin gerekse hücrelerarası bağlantılara bağlı bir telafi mekanizması çalışmaktadır. İkincisi, epididimiste gelişen seminal granülomu seminifer tübül iç basıncını artırarak adlüminal kompartımanda spermatid-Sertoli hücresi ilişkisini bozup spermatidlerin olgunlaşamadan hızla dejenere olmasına neden olabilir.

The effects on the testis of open-ended vasectomy in the Wistar rat have been assessed morphometrically and evaluated statistically at 1, 4, 12, and 24 weeks after operation, and compared to those in sham-operated controls. The open-ended technique, ie, without ligation of the epididymal end of the cut ductus deferens, allowed drainage of sperm into the spermatic cord. During the period of our study, it did not affect the structure of the testis; although the germinal epithelium, basement membrane, and the stroma and cells of the interstitial space all showed variations when compared to the control group, these were not statistically significant.
