Definition of the minimal viral components required for the initiation of unprimed RNA synthesis by influenza virus RNA polymerase

Abstract: The first 11 nt at the 5' end of influenza virus genomic RNA were shown to be both necessary and sufficient for specific binding by the influenza virus polymerase. A novel in vitro transcription assay, in which the polymerase was bound to paramagnetic beads via a biotinylated 5'-vRNA oligonucleotide, was used to study the activities of different forms of the polymerase. Complexes composed of co-expressed PB1/PB2/PA proteins and a sub-complex composed of PB1/PA bound to the 5'-vRNA oligonucleotide, whereas PB1 … Show more

“…Although PB1 alone can bind the 5Ј-terminal vRNA sequence (10), its binding is more efficient upon PB1-PA complex formation (20), suggesting a role for PA in the ability of the polymerase to interact with the vRNA 5Ј terminus. A polymerase complex with alterations in the RNA binding properties could be unsuitable for cRNA synthesis.…”

“…95 Although it has been reported that alternative binary complexes of the polymerase subunits might be responsible for transcription or replication, 120,121 a number of other reports indicate that the polymerase heterotrimer is the functional unit in both processes. 8,48,49,63,66,122 Altogether, the genetic approaches indicated above enabled the proposal of a new model for influenza RNA replication, which is depicted in the diagram presented in Figure 5. Activation of vRNA synthesis would result from the interaction of a soluble polymerase complex (Non-resident polymerase-NRP1-) with the cRNP (Fig.…”
