Defining candidate mRNA and protein EV biomarkers to discriminate ccRCC and pRCC from non-malignant renal cells in vitro

Abstract: Renal cell carcinoma (RCC) accounts for over 400,000 new cases and 175,000 deaths annually. Diagnostic RCC biomarkers may prevent overtreatment in patients with early disease. Extracellular vesicles (EVs) are a promising source of RCC biomarkers because EVs carry proteins and messenger RNA (mRNA) among other biomolecules. We aimed to identify biomarkers and assess biological functions of EV cargo from clear cell RCC (ccRCC), papillary RCC (pRCC), and benign kidney cell lines. EVs were enriched from conditioned… Show more

“…A total of 1,726 proteins and 461 mRNAs were quantified in EVs derived from clear cell renal cell carcinoma, papillary renal cell carcinoma, and benign kidney cell lines. Among them, 181 proteins and 159 mRNAs were detected in all samples (Zieren et al, 2021). Shared proteins represent factors involved in biogenesis, whereas unique proteins reflected the characteristics of their cell of origin and were therefore proposed as biomarkers.…”

“…For example, Zieren et al performed a multi-omic analysis on RCC and benign kidney cell lines. They identified 34 proteins as a candidate for pRCC biomarkers in EVs, and 20 proteins and 8 mRNA for ccRCC [ 90 ]. The results of this basic research pushed them to verification of their hypothesis in a human study.…”
