Deficiency of the suppressor inducer subset of T lymphocytes in rheumatoid arthritis

Abstract: New monoclonal antibodies allow CD4 lymphocytes to be categorized into helperhnducer (HI) CD4+ 4B4-t, and suppressor inducer (SI) CD4+ 2H4+ subpopulations. Blood and synovial lymphocytes from 30 patients with rheumatoid arthritis (RA), 13 patients with other articular diseases, and 24 control subjects were exposed to these monoclonal antibodies and analyzed by flow cytometry. CD4:CDS ratios were similar for the 3 groups. In RA patients, there was a depletion of SI cells in blood and synovial fluid, and the rat… Show more

“…PB and SF T cells from the same RA patient and PB T cells from a normal donor were stained with various MAb and analyzed (Figure 1). In a total of 5 experiments, the distribution of CD4+, CD8+, CD45RA+, and CD45RO+ T cells was comparable with that previously reported (1, 19,20 Thus, SF and ST are enriched in T cells expressing CD45R0, that are CD45RBdi". There were no differences in CD45R exon expression between normal and RA PB.…”

“…For this purpose, the analysis of the expression of the isoforms of CD45 is of particular value. Synovial T cells are known to be enriched in CD45RO+ memory T cells compared with PB T cells (19,20). Memory T cells in humans can now be defined more completely by MAb that recognize the products of the B exon of the CD45 gene.…”

“…The naive subset has been shown to be greatly reduced in RA synovial fluid (18,19) and in peripheral blood (18). Thus far, despite intensive but conflicting investigations, no definite functional subpopulation has been distinguished.…”
