Deeper Understanding of Biological Tissue: Quantitative Correlation of MALDI-TOF and Raman Imaging

Abstract: In order to achieve a comprehensive description of biological tissue, spectral information about proteins, lipids, nucleic acids, and other biochemical components need to be obtained concurrently. Different analytical techniques may be combined to record complementary information of the same sample. Established techniques, which can be utilized to elucidate the biochemistry of tissue samples are, for instance, MALDI-TOF-MS and Raman microscopic imaging. With this contribution, we combine these two techniques f… Show more

“…More recently, Fagerer et al 19 combined single cell fluorescence and Raman imaging with MALDI mass spectrometry (MS) profiling of algae in order to visualize secondary metabolite production and the associated depletion of cellular adenosine triphosphate, impressively demonstrating how combining these techniques can yield a more comprehensive biological picture. Finally, a landmark-based histology registration strategy was developed for semiautomated alignment between MALDI-time-of-flight (TOF) and Raman imaging platforms by Bocklitz et al 20 …”

“…187 A number of investigators have also recently combined confocal Raman microscopy and IMS to provide better characterization of tissue differentiation and disease state alterations. 188190 The use of multiple modalities can also enable cross-platform interpretation and/or validation of results, such as using MALDI spectra to help unravel complex Raman spectra from tissue specimens. 191 So-called image fusion approaches, where datasets from different imaging experiments (e.g., microscopy and IMS) are mathematically combined, offer methods for exploiting data from multimodal experiments.…”

“…Raman microspectroscopy has been combined with nanoscale secondary ion mass spectrometry (NanoSIMS) to quantify the bacterial uptake of deuterium during heavy water labeling experiments (Berry et al, 2015). Moreover, matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-ToF-MS) is compatible with microbiological analyses and Raman imaging (Bocklitz et al, 2013; Pande et al, 2016; Stasulli and Shank, 2016). Although it has not yet been applied for the imaging of cells within complex environments such as soils, MALDI-ToF-MS been used to characterize individual bacterial colonies (Pande et al, 2016; Stasulli and Shank, 2016).…”
