Deep eutectic solvents as novel extraction media for protein partitioning

Abstract: Four kinds of green deep eutectic solvent (DES) were synthesized, including choline chloride (ChCl)-urea, tetramethylammonium chloride (TMACl)-urea, tetrapropylammonium bromide (TPMBr)-urea and ChCl-methylurea. An aqueous two-phase system (ATPS) based ChCl-urea DES was studied for the first time for the extraction of bovine serum albumin (BSA). Single factor experiments proved that the extraction efficiency of BSA was influenced by the mass of the DES, concentration of K2HPO4 solution, separation time and extr… Show more

“…Among these solvents, ionic liquids (ILs) have generated greater attention recently in the global scientific communities with the aim of developing ecofriendly solvents, because ILs possess attractive properties such as non-volatility (Grasvik et al 2014) and tailor-made reagents for selective extractions (Xia et al 2014;Zeng et al 2014) with lower melting points, in general, below 100°C which makes easy handling of IL solvents. However, synthesis of ILs is a cost-intensive process due to the high production costs of the individual IL components.…”

“…16 Albeit scarcely investigated, the potential of DES as phase forming components of ABS was recently reported by Xu and co workers. 17,18 Yet, those pioneering phenomenological results did not comprise the mechanisms associated with the coexisting phases demixing and the stability of the HBD:HBA complex in aqueous media. Only "ternary" phase diagrams for each ABS were reported, and their applications to the extraction of proteins were presented.…”

“…Only "ternary" phase diagrams for each ABS were reported, and their applications to the extraction of proteins were presented. 17,18 Whether DES in the form of a HBD:HBA complex will form a ternary system, as in common ABS, or the alleged DES based ABS is a quaternary system that phase separates with a nonstoichiometric partition of the HBD and HBA species between the two phases remains an open question. In this context, the present work aims at shedding light on this subject.…”

“…DLLME-SFO is becoming a popular sample preparation technique, and the type of extraction solvents has been developed in recent years, e.g. [15] and has gradually been considered a new type of green ionic liquid [16][17][18][19]. DESs are composed of two or three components with a melting point lower than its individual components, consisting of hydrogen bond donor (HBD), and hydrogen bond acceptor (HBA) molecules in a certain molar ratio [20].…”
