Decreased TMPRSS2 expression by SARS-CoV-2 predicts the poor prognosis of lung cancer patients through metabolic pathways and immune infiltration

Abstract: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has rapidly spread around the world and became a global pandemic in 2020. One promising drug target for SARS-CoV-2 is the transmembrane protease serine 2 (TMPRSS2). This study was designed to explore the expression status, prognostic significance and molecular functions of TMPRSS2 in lung cancer. TMPRSS2 expression was investigated using the TIMER, Oncomine, UALCAN, GEO, HPA and TCGA databases. The prognostic value of TMPRSS2 was examined using Cox r… Show more

“…Because SARS-CoV infection downregulates TMPRSS2 expression in cultured cells [ 22 ], we investigated the changes in TMPRSS2 expression in COVID-19-infected lungs. By analyzing the single-cell RNA-sequencing dataset (GSE171524) of COVID-19-infected lungs and control lungs [ 23 ], the expression levels of TMPRSS2 in COVID-19-infected lungs were significantly reduced compared to the control lungs ( Figure 7 A).…”

“…Immunohistochemical (IHC) staining results were directly obtained from the HPA (Human Protein Atlas) database ( https://www.proteinatlas.org/ ) as described previously [ 36 , 87 ]. The protein levels of PRGs in normal liver tissues and HCC tissues were compared through IHC staining.…”

“…For example, the TMPRSS2 gene expression was significantly reduced in patients with LUAD as compared with healthy patients, although the ACE2 expression was higher in the former group ( 84 86 ). Moreover, the TMPRSS2 gene expression was significantly reduced in patients with LUSC as compared with healthy patients, although the ACE2 expression was high in the patients with both LUAD and LUSC ( 87 ).…”
