Decreased Taurine and Creatine in the Thalamus May Relate to Behavioral Impairments in Ethanol-Fed Mice: A Pilot Study of Proton Magnetic Resonance Spectroscopy

Abstract: Minimal hepatic encephalopathy (MHE) is highly prevalent, observed in up to 80% of patients with liver dysfunction. Minimal hepatic encephalopathy is defined as hepatic encephalopathy with cognitive deficits and no grossly evident neurologic abnormalities. Clinical management may be delayed due to the lack of in vivo quantitative methods needed to reveal changes in brain neurobiochemical biomarkers. To gain insight into the development of alcoholic liver disease–induced neurological dysfunction (NDF), a mouse … Show more

“…In vivo magnetic resonance spectroscopy measurement of ethanol metabolites. In vivo MRS experiments were performed on a Bruker BioSpec 70/30USR Advance III 7T horizontal bore MR scanner (Bruker Biospin MRI) as described previously 65,66 . All mice were weighed, anaesthetized and placed in a prone position on a specially designed cradle and inserted into the MR scanner for a baseline measurement.…”

“…One of the criteria for a successful animal model of alcoholism is the demonstration of physical dependence per se must be unequivocally attributed to the chronic administration and subsequent removal of ethanol. Common models to induce physical dependence to alcohol (cf., Mello, 1973) include exposure to ethanol via drinking (e.g., Xu et al, 2018), intraperitoneal (i.p.) (e.g., Liew et al, 2016) or intragastric (i.g.)…”

They use metabolite ratios rather than absolute concentrations, which has been challenged since tCr could have abnormal levels (23,24) and thus, it is hard to draw definite conclusions on which of the two metabolites in the ratio was responsible for the distinction. There are some reports of tCr values changing across the MCI-AD spectrum (25,26).…”
