Decreased expression of protooncogenes c-fos, c-myc, and c-jun following polyamine depletion in IEC-6 cells

Abstract: Direct exposure of small intestinal mucosal cells to luminal polyamines stimulates proliferation. This study tests the hypothesis that the protooncogenes c-fos, c-myc, c-jun, and junB are involved in the mechanism by which polyamines modulate mucosal growth. Studies were conducted in the IEC-6 cell line, derived from rat small intestinal crypt cells. Cells were grown in Dulbecco's minimal essential medium containing 5% dialyzed fetal bovine serum (dFBS) in the presence of absence of alpha-difluoromethylornithi… Show more

“…The focus of the current study went further to determine the involvement of the Akt signaling pathway in this process. Consistent with our previous studies (10,36), inhibition of ornithine decarboxylase activity by exposure to 5 mM DFMO almost completely depleted cellular polyamines in IEC-6 cells. Putrescine was undetectable on day 2 after treatment with DFMO.…”

“…ODC is the enzyme responsible for catalyzing the first rate-limiting step in polyamine biosynthesis. Inhibition of ODC activity by DFMO decreases mRNA levels for the c-fos, c-myc, and c-jun proto-oncogenes in IEC-6 cells (18). In the present report we investigated the potential role of the NF-B transcription factor family in the regulation of gene expression by polyamines.…”

“…Although the molecular mechanisms whereby polyamines control the growth of intestinal mucosal cells have not been fully elucidated, they are known to be involved in the regulation of growth-related genes (8,9,13,32,33). In this regard, polyamines have been shown to suppress the expression of growthinhibiting genes such as p53 (10,12), NPM (14), p21 (34), TGF␤/TGF␤-Rs (TGF␤-receptors) (8,35), and junD (11,39), primarily by controlling mRNA stability.…”
