Decreased Expression of Leptin among Patients with Shoulder Stiffness

Abstract: Shoulder stiffness (SS) is a disease that is fibroblastic and inflammatory in nature. Leptin is an adipokine-mediating the fibroblastic and inflammatory processes of various diseases. Our study tried to investigate the role of leptin in SS pathogenesis. Subacromial bursa from stiff and non-stiff shoulders were obtained for reverse transcription-polymerase chain reaction (RT-PCR) analysis and immunoblotting. Subacromial fluid was obtained for enzyme-linked immunosorbent assay. We showed that the expression leve… Show more

“…The current study demonstrated that leptin expression was increased in capsule tissue of FS, suggesting that leptin might participate in the inflammatory and fibroblastic processes of FS, similar to its role in other fibrotic diseases. In contrast, Ko et al 25 showed that the expression of leptin was lower in the subacromial bursae and fluid of stiff shoulders. These findings appear to contradict the proinflammatory nature of leptin and the results of other fibrotic disorders.…”

“…The assessment of gene transcription levels was conducted employing the reverse transcription-quantitative polymerase chain reaction (RT-qPCR) assay following our previous works (Ko et al 2022a , b , 2023a , b ). Tissues were pulverized under liquid nitrogen in the absence of RNase, and total RNA was extracted using the RiboPure RNA purification kit (Thermo Fisher Scientific, Waltham, MA, USA).…”
