Decreased Anti-Müllerian hormone and Anti-Müllerian hormone receptor type 2 in hypothalami of old Japanese Black cows

Abstract: Cow fertility decreases with age, but the hypothalamic pathomechanisms are not understood. Anti-Müllerian hormone (AMH) stimulates gonadotropin-releasing hormone (GnRH) neurons via AMH receptor type 2 (AMHR2), and most GnRH neurons in the preoptic area (POA), arcuate nucleus (ARC), and median eminence (ME) express AMH and AMHR2. Therefore, we hypothesized that both protein amounts would differ in the anterior hypothalamus (containing the POA) and posterior hypothalamus (containing the ARC and ME) between young… Show more

“…We obtained brain samples (cortex, hippocampus, anterior hypothalamus containing the SCN [29][30][31] and in Supplementary Figure 1. The frontal lobe cortex was collected caudal to the central sulcus near the midline.…”

“…We used a previously reported method, with minor modifications, for western blotting of bovine brain [30,31]. Briefly, proteins were extracted from the frozen stock tissues of young and old groups (n = 6 per region per group) using a tissue protein extraction reagent (T-PER; Thermo Fisher Scientific) with a Halt Protease Inhibitor Cocktail (Thermo Fisher Scientific).…”

“…Old cows were slaughtered after parturition a sufficient number of times as planned by farmers to obtain beef, usually after 84 months of age. We obtained whole brains (including the hypothalamus) for EPl extraction from healthy, post-pubertal, young Japanese Black heifers (2.2 ± 0.0 years old, n = 6) and old Japanese Black cows (10.3 ± 1.6 years old, n = 6) from the local abattoir, and we preserved these without air at À80 C. We obtained hypothalami samples for RNAseq from different healthy, post-pubertal, non-pregnant, non-lactating Japanese Black heifers (2.2 ± 0.0 years old, n = 5) and from old Japanese Black cows (10.3 ± 1.6 years old; 7.4 ± 1.1 parity; n = 5), according to the method reported previously (Kereilwe & Kadokawa, 2020a). Briefly, brain tissues were dissected as follows: rostral border of the optic chiasm, caudally rostral to the mammillary bodies, lateral to the optic chiasm, and 0.5 cm dorsal to the third ventricle.…”
