Decorin as a multivalent therapeutic agent against cancer

Abstract: Decorin is a prototypical small leucine-rich proteoglycan and epitomizes the multifunctional nature of this critical gene family. Soluble decorin engages multiple receptor tyrosine kinases within the target rich environment of the tumor stroma and tumor parenchyma. Upon receptor binding, decorin initiates signaling pathways within endothelial cells downstream of VEGFR2 that ultimately culminate in a Peg3/Beclin 1/LC3-dependent autophagic program. Concomitant with autophagic induction, decorin blunts capillary … Show more

“…Decorin (Dcn) is a SLRP that is most closely related to Bgn (6), yet in contrast to what we found for Bgn, Dcn has inhibitory effects on angiogenesis (25,26). It is unclear why Dcn and Bgn have what appears to be opposite effects on angiogenesis, but it is likely that tissue context is involved (27).…”

“…Decorin controls also the bioavailability of TGF-β whose release depends on the activity of different proteases involved in angiogenesis [166,167]. Furthermore, soluble decorin is a high affinity antagonistic ligand for several key receptors tyrosine kinases including Epithelial Growth Factor Receptor (EGFR), HGF Receptor, IGF-1 Receptor and VEGFR2 [168]. The engagement of decorin with cell surface receptors can either activate or inhibit the function of the receptor, depending on the physiological state of the tissue [169].…”

“…Decorin, a component of connective tissue, binds to collagen and plays an important role in matrix assembly. Furthermore, decorin, like elastin, is capable of suppressing the growth of various tumor cell lines [73, 74]. Interestingly, with the reduction of versican expression, a number of sulfo-, acetylgalactosaminyl-, and xylotransferases associated with the biosynthesis of versican and other proteoglycans were elevated, while others were reduced.…”
