Decay Accelerating Factor Can Control T Cell Differentiation into IFN-γ-Producing Effector Cells via Regulating Local C5a-Induced IL-12 Production

Abstract: A newly recognized link between the complement system and adaptive immunity is that decay accelerating factor (DAF), a cell surface C3/C5 convertase regulator, exerts control over T cell responses. Extending these results, we show that cultures of Marilyn TCR-transgenic T cells stimulated with DAF-deficient (Daf1−/−) APCs produce significantly more IL-12, C5a, and IFN-γ compared with cultures containing wild-type APCs. DAF-regulated IL-12 production and subsequent T cell differentiation into IFN-γ-producing ef… Show more

“…Donor spleen cell stimulators were treated with mitomycin C to limit proliferation and cytokine secretion. 18 After washing, detection antibodies (BD Biosciences) were added overnight. After washing, alkaline phosphatase-conjugated anti-biotin antibody (Vector Laboratories) diluted 1:2000 in PBS Tween 1% bovine serum albumin was added for 90 minutes, the plates were developed, and the resulting spots were counted on an ImmunoSpot Series 4 Analyzer (Cellular Technology Ltd., Shaker Heights, OH).…”

“…3,4, 8 Lalli et al 4 argue that, in the absence of Daf1, complement activation on antigen-presenting cells (APCs) leads to IL-12 production and a T helper type 1 (Th1), IFN-c, driven CD4…”

“…2 Furthermore, it has been shown in a variety of mouse autoimmune models that Daf1 plays a protective role in autoimmune disease. [3][4][5][6][7] It has been argued that Daf1 exerts control over T-cell responsiveness in a complement-dependent manner, influencing the expression of interferon (IFN)-c and interleukin (IL)-12.…”

“…11 The generation of C5a locally at the site of interaction between antigen-presenting cells and T cells provides a survival signal for T cells 12 and is required for development of effector functions. 13 This is a significant effect and is sufficient to modify the survival of murine cardiac allografts. 14 In the study by Gueler et al, 4 blocking C5a receptor signaling at a time when an allo-specific T cell response is developing is sufficient to prevent later rejection.…”
