Death of ouabain-treated renal epithelial cells: evidence for p38 MAPK-mediated Na i + /K i + -independent signaling

Abstract: Recent studies demonstrate that cytotoxic actions of ouabain and other cardiotonic steroids (CTS) on renal epithelial cells (REC) are triggered by their interaction with the Na(+),K(+)-ATPase alpha-subunit but not the result of inhibition of Na(+),K(+)-ATPase-mediated ion fluxes and inversion of the [Na(+)](i)/[K(+)](i) ratio. This study examined the role of mitogen-activated protein kinases (MAPK) in the death of ouabain-treated REC. Exposure of C7-MDCK cells that resembled principal cells from canine kidney … Show more

“…4b). This small portion of a1R subunits maintains cell survival via their interactions with an unknown adaptor protein III that, in turn, inhibits p38 MAPK-mediated signaling cascade documented in our recent investigations [43]. This model is consistent with cytotoxic action of 1000 lM ouabain in a1R-C7 cells seen in K ?…”

“…Several lines of evidence have demonstrated that the effect of ouabain on p38 MAPK phosphorylation is cell-type specific [36,55]. We observed here that ouabain was able to induce p38 MAPK activation in lung epithelial cells.…”

“…They examined several signaling pathways, including PI3K, MAPK ERK1/2 kinase, tyrosine kinases and Ras signaling, in ouabain-treated renal epithelial cells, but none of the signaling systems tested affected ouabain-induced cell death [11]. However, in a more recent study, Akimova et al [12] indicated that p38 MAPK signaling was involved in ouabain-induced renal epithelial cell death but not in HUVEC. In spite of this extensive research, the exact mechanism of ouabain-induced cell death is not fully understood.…”
