DEAD box RNA helicases crucial regulators of gene expression and oncogenesis

Abstract: DEAD box protein family of RNA helicases are vital players of RNA metabolism, and constitute the largest family of RNA helicases. Members of this family share nine conserved motifs including an Asp-Glu-Ala-Asp motif, giving this family its characteristic name as DEAD box RNA helicases. These conserved motifs confer RNA binding and RNA unwinding properties. Besides functioning in RNA metabolism, emerging evidences suggests several DEAD box RNA helicases to possess potential roles in regulating gene expression b… Show more

“…DDX43 has published ATP-dependent RNA helicase activity (Talwar et al, 2017), whereas DDX53 has not been biochemically characterized to date. Similar to DDX5 and DDX17, DDX43 and DDX53 are linked to cancers (Sarkar & Ghosh, 2016). To permit genetic analysis of DDX5 function, Iggo et al cloned yeast genes related to DDX5 and named them dbp2 in Schizosaccharomyces pombe and DBP2 in Saccharomyces cerevisiae ( S. cerevisiae ), for DEAD-box protein 2 (Iggo et al, 1991).…”

“…Mounting evidences suggest p68 to play a crucial role in cancer development and progression. It is reported to be overexpressed in various cancers and acts as a transcriptional co-activator of several transcription factors like β-catenin, NF-κB p50, MAML1 and Androgen Receptor [14].…”

“…The cellular superfamily of RNA helicases, also known as DExD/H-box helicases, are involved in every aspect of RNA metabolism 15 , 16 . However, in recent years, their involvement has been identified in an increasing number of other cellular functions such as: innate immune responses against pathogens, oncogenesis, and inflammatory diseases 17 , 18 .…”
