De novo CD5-positive diffuse large B-cell lymphomas show high specificity for cyclin D2 expression

Abstract: D cyclins positively regulate the cell cycle and mediate the pathogenesis of some lymphomas. Cyclin D1 overexpression is the hallmark of mantle cell lymphoma, whereas cyclins D2 and D3 are reportedly not as specific to certain lymphomas as cyclin D1. In this study, cyclin D2 was found to be overexpressed in 98% of de novo CD5-positive diffuse large B-cell lymphomas (DLBCLs) (50/51) and in 28% of CD5-negative DLBCLs (14/51). A statistically significant difference was observed between these two groups (p<0.0001)… Show more

“…Overexpression of cyclin D2 was also found to be associated with CD5 positivity in DLBCL, especially in de novo cases (24). In our cohort, the low number of cyclin D2-positive cases also did not allow statistical analysis.…”

“…Expression of cyclin D2 has been also repeatedly studied in DLBCL. The fraction of cyclin D2-positive cases varies between 13 and 62% and this feature was shown to be an independent indicator of poor survival (18,(22)(23)(24). Cyclin D2 overexpression is closely associated with CD5-positive cases de novo (24).…”

“…Although there is no cytomorphologic difference between CD5 þ and CD5 À DLBCL NOS, cyclin D2 has been reported to be highly specific for de novo CD5 þ DLBCL. 30 For practical diagnostics, the pleomorphic variant of MCL and the DLBCL variant of the Richter transformation from CLL/SLL should be ruled out in CD5 þ B-cell lymphomas with medium to large cell sizes. The CD5…”
