De novo assembly and characterization of the transcriptome in the desiccation-tolerant moss Syntrichia caninervis

Abstract: BackgroundSyntrichia caninervis is a desiccation-tolerant moss and the dominant bryophyte of the Biological Soil Crusts (BSCs) found in the Mojave and Gurbantunggut deserts. Next generation high throughput sequencing technologies offer an efficient and economic choice for characterizing non-model organism transcriptomes with little or no prior molecular information available.ResultsIn this study, we employed next generation, high-throughput, Illumina RNA-Seq to analyze the poly-(A) + mRNA from hydrated, dehydr… Show more

“…This moss has evolved a constitutive protection system to alleviate the damage caused by dehydration (Oliver et al 2000, 2009). Syntrichia caninervis and Bryum argenteum are both classified as DT mosses, and their responses to desiccation have been widely studied (Gao et al 2014, 2015; Li et al 2014; Wu et al 2012; Zheng et al 2011). …”

“…We identified 26 Unigenes as ScELIPs in the reference transcriptome [10], 6of which have complete open reading frames (ORFs): Unigenes 13021, 40120, 40121, 68225, 16576, and 8044. Unigene 16576 and 8044 each contain 94 predicted amino acids that were of short length to known ELIPs (Table S1).…”

“…Publicly available ELIP sequences of A. thaliana and Physcomitrella patens were used to screen the transcriptome of S. caninervis [10] for single copy transcripts (designated as unigenes) that were putative ELIP coding sequences. We translated each unigene to obtain the amino acid sequence using ORF finder.…”

“…In the OneKP 71 pilot study, transcriptome data for a total of 92 plants were generated and among them 11 moss species were included 72 (Wickett et al, 2014). The transcriptomes in two desert mosses Syntrichia caninervis and Bryum argenteum were reported in 73 our previous studies (Gao et al, 2014;Gao et al, 2017) and a recent phylotranscriptomic study in pleurocarpous mosses 74 (Hypnales) generated a large set of transcriptomic data . 75…”
