Ddx56 maintains proliferation of mouse embryonic stem cells via ribosome assembly and interaction with the Oct4/Sox2 complex

Abstract: Background: Embryonic stem cells (ESCs) are important source of clinical stem cells for therapy, so dissecting the functional gene regulatory network involved in their self-renewal and proliferation is an urgent task. We previously reported that Ddx56 interacts with the core transcriptional factor Oct4 by mass spectrometry analysis in ESCs. However, the exact function of Ddx56 in ESCs remains unclear. Methods: We investigated the role of Ddx56 in mouse ESCs (mESCs) through both gain-and loss-of-function strate… Show more

“…DDX56 is a component of free nucleoplasm 65S preribosomal particles, which may participate in the assembly of the 60S ribosomal subunit. DDX56 has also been reported to regulate ribosome assembly through its association with the Oct4 and Sox2 complex, which is necessary to maintain embryonic stem cells (ESCs) proliferation [ 91 ]. DDX56 is upregulated in osteosarcoma and colorectal cancer ( Table 1 ) [ 92 , 93 , 94 ].…”

“…DDX56 is located on chromosome 7 and regulates diverse RNA metabolic processes, including transcription, translation, ribosome assembly, ribosome maturation, pre-mRNA splicing, and mRNA degradation [ 7 , 35 ]. According to results of EdU and CCK8 assays, knockdown of DDX56 suppressed GC cell proliferation, while overexpression of DDX56 promoted proliferation.…”

“…DDX56 is a nucleolar protein and participates in ribosome biogenesis and ribosome assembly (68,69). Furthermore, DDX56 is involved in transcription (70), pre-mRNA splicing (71), as well as miRNA-mediated post-transcriptional regulation by degradation of primary miRNA (72).…”
