DDM1 guards against telomere truncation in Arabidopsis

Abstract: Epigenetic pathways, including DNA methylation, are crucial for telomere maintenance. Deficient in DNA Methylation 1 (DDM1) encodes a nucleosome remodeling protein, required to maintain DNA methylation in Arabidopsis thaliana. Plants lacking DDM1 can be self-propagated, but in the sixth generation (G6) hypomethylation leads to rampant transposon activation and infertility. Here we examine the role of DDM1 in telomere length homeostasis through a longitudinal study of successive generations of ddm1-2 mutants. W… Show more

“…Very low level of genomic DNA methylation caused disturbances in telomere homeostasis in A. thaliana ( Ogrocká et al 2014 , Xie and Shippen 2018 ), while no such changes were observed in N. tabacum ( Majerová et al 2011 ). This shows the differences in the role of DNA methylation in the regulation of telomere homeostasis in various plants ( Fojtová and Fajkus 2014 , Procházková-Schrumpfová et al 2019 ).…”

“…This pattern was confirmed by an independent approach, with high reliability at least in the proximal part of the telomere [203,204], and methylated telomeric cytosines were detected in cultured Nicotiana tabacum (tobacco) cells [205] and other plants [206]. Disruption of telomere homeostasis as a consequence of decreased genomic DNA methylation was observed in A. thaliana [203,207] but not in tobacco cells [205], which shows differences in the involvement of DNA methylation in regulation of telomere homeostasis between these model plants (for a more detailed review see Reference [208]).…”

“…The short telomere phenotype and hypersensitivity to the induction of DNA double-strand breaks were previously observed in Arabidopsis ddm1 mutants. This correlation raised the hypothesis that genome hypomethylation-induced increase in HR and stochastic deletional recombination led to telomere shortening [ 29 ]. Intermolecular HR has been reported to be a dominant mechanism for detoxification of zebularine-induced DNA damage, independently of changes in DNA methylation [ 18 ].…”
