Day/night and circadian rhythm control of con gene expression in Neurospora.

Abstract: In the filamentous fungus Neurospora crassa, several events in the process of conidiation are influenced by light. Two genes, con-6 and con-10, which were previously shown to be transcriptionally activated during conidiation and by exposure to light, were found to be unexpressed in mycelium maintained in constant darkness or in constant light. However, when mycelium was shifted from darkness to light, transcripts of both genes appeared and were abundant. Upon further illumination both transcripts disappeared--… Show more

“…However, the amount of con-10 mRNA that accumulated in light-exposed mycelia was increased in strains with mutations in acon-3, fl, and fld, which suggested that the corresponding gene products should repress con-10 photoactivation ( Figure 4A). The increase in the accumulation of con-10 mRNA after light exposure in these developmental mutants had been reported after RNA hybridization experiments (Lauter and Yanofsky 1993), further supporting our results.…”

“…In addition, we assayed light-dependent fl mRNA accumulation in different fl mutants. As a control, we assayed the light-dependent accumulation of con-10 mRNA since light regulation for this gene has been observed in several developmental mutants (Lauter and Yanofsky 1993). We observed that fl was activated by light in all the developmental mutants that we tested, with the exception of fld ( Figure 4A).…”

“…Photoadaptation has been described for light-regulated genes in N. crassa (Lauter and Yanofsky 1993;Arpaia et al 1999;Schwerdtfeger and Linden 2003) and P. blakesleeanus (Rodriguez-Romero and Corrochano 2006). Photoadaptation in N. crassa requires the product of the vvd gene (Heintzen et al 2001;Schwerdtfeger and Linden 2003;Zoltowski et al 2007), such that the activity of the white-collar complex (WCC) is controlled by the VVD protein through the direct interaction of VVD with the components of the WCC (Chen et al 2010a;Hunt et al 2010;Malzahn et al 2010).…”

“…The con-6 and con-10 proteins are present in a second type of asexual spore, the microconidium (10,13), and the con-10 protein is present in sexual spores (10, 13). Expression of con-6 and con-10 can also be induced in vegetative mycelial cultures by light exposure or by imposing conditions that induce circadian rhythm (10,(13)(14)(15). We describe here the selection and partial characterization of mutants that express con-6 and con-10 aberrantly during vegetative mycelial growth.…”
