New emerging entomopathogenic fungi isolated from soil in South Sumatra (Indonesia) and their filtrate and conidial insecticidal activity against Spodoptera litura. Biodiversitas 21: 5102-5113. This study was carried out to confirm identity of Beauveria bassiana and Metarhizium anisopliae using sequence analysis of Internal Transcribed Spacer (ITS) region as well as the two unknown isolates of EPF and to investigate their filtrate and conidial insecticidal activity against S. litura larvae. The EPF species were identified based on morphological characters and molecular analysis. Their culture filtrate and conidial bioassays were performed on S. litura larvae. The species found were Penicillium citrinum (BKbTp), Talaromyces diversus (MSwTp1), B. bassiana (BSwTd4), and M. anisopliae (MSwTp3) and have been deposited in the GenBank. All EPF species have high insecticidal activity against S. litura larvae. The highest larvae mortality caused by filtrate was found on P. citrinum (98.67%) and not significantly different from those of T. diversus (96%), but significantly different from B. bassiana (86.67%) and M anisopliae (82.67%). The highest larvae mortality caused by the conidia was also occurred to P. citrinum (98.67%) which was not significantly different from those of T. diversus (97.33%), B. bassiana (86.67%), and M. anisopliae (86.67%). The LT50 caused by T. diversus filtrate was the shortest (6.19 days) and not significantly different from those caused by P. citrinum (6.62 days). The shortest LT50 caused by conidia was found on P. citrinum (5.45 days). Therefore, the conidia and filtrate of P. citrinum, a new emerging entomopathogenic fungus, has the highest insecticidal activity. P. citrinum has the potential to be developed into commercial mycoinsecticides.

The response of chili (Capsicum annuum L.) to oriental fruit fly infestation (Bactrocera dorsalis) is highly variable among varieties. The differences in the resistance level of chili to oriental fruit fly infestation are presumed to be determined by the characteristics of chili fruit. This study aims to evaluate the morphochemical characteristics of different resistance levels of chili fruits to oriental fruit fly infestation in field conditions. The field test was carried out at the research station of the Indonesian Vegetable Research Institute (IVEGRI), West Java, Indonesia. Six essential derivatives of C. annuum from IVEGRI, consisting of three resistant and three susceptible varieties, were established in a prior investigation. The test population included 132 plants, with 22 plants planted for each variety. The resistance parameters observed were oviposition incidence, yield loss, fitness index, and chili fruit characteristics (morphology, nutrition, volatile compounds). The results showed that there were morphological and chemical differences between the varieties resistant and susceptible to oriental fruit fly infestation. The morphological characteristics of the fruit (width, weight, and thickness of fruit flesh) and fruit shape at pedicel attachment had an impact on the resistance level of fruit flies. Meanwhile, volatile compounds, water content, carbohydrates, and fiber content were among the chemical features that influenced oriental fruit fly infestation.

Ovipositions of fruit flies typically leave some wounds on the chili fruits. These punctures might allow some pathogenic microorganisms to get into the fruits and eventually cause the secondary infections. Taking this into account, this study aimed at determining the expression of chili defense gene (CaRGA2) due to pathogenic fungus infection which occurred following the fruit fly oviposition activities. The research was carried out from April to October 2018 in two research facilities located in West Java, Indonesia, i.e. the Molecular Laboratory of IVEGRI (West Bandung) and the Molecular Biology Laboratory of the Biotechnology and Bioindustry Research Center (Bogor). Chili fruits of three chili varieties with different levels of resistance againts fruit fly infestations (i.e. susceptible, moderately resistant, and resistant) were used in this study. Moreover, the RNA of each of the varieties was isolated before and after the fruit fly oviposition. The chili defense gene of CaRGA2 was tested in this study whereas the housekeeping gene used was Actin. The gene was amplified by using the RT-PCR method and afterwards, the data from the amplification were analyzed by employing the Livak method. The results showed that chili defense gene expression increased significantly in the susceptible chili variety, but did otherwise in both the moderately resistant and resistant varieties. Eventually, this result supported the previous studies that revealed the increased fungal pathogen infections in host plants due to the injuries caused by fruit fly oviposition.
