Dataset of growth cone-enriched lipidome and proteome of embryonic to early postnatal mouse brain

Abstract: A growth cone is a part of a neuron considered as a hub for axon growth, motility and guidance functions. Growth cones are thought to play a critical role during development of neurons. Growth cones also play a significant role in adult regeneration. Here, we present a dataset on the lipid and protein profiling of the growth cone-enriched fractions derived from C57BL/6J mice forebrains of developmental stage: E18, P0, P3, P6, and P9. For comparison, we analyzed non-growth cone membranes from the same samples. … Show more

“…Lipid synthesis and transport are main components for plasmalemma expansion (Pfenninger, 2009; Pfenninger et al, 2003; Pfrieger & Ungerer, 2011). A comprehensive analysis of growth cones (Trzeciecka & Bhattacharya, 2019) and multiple models of regenerating optic nerve (Trzeciecka, Carmy, et al, 2019; Trzeciecka, Stark, et al, 2019) are now available to help identify these critical molecules. Various innovative approaches are emerging which may be adapted for reinnervation after long distance directed axon regeneration.…”

“…The peptides were analyzed using a Q Exactive mass spectrometer (Thermo Fisher Scientific) equipped with a heated electrospray ionization source (HESI) operating in positive ion mode, as described previously (Trzeciecka & Bhattacharya, 2019). The spray voltage was 1.8 kV, all gases were set to 0, the capillary temperature was 250 ˚C and the S-lens radio frequency level was 50.…”

“…The data was searched against Mus musculus entries in the Uniprot protein sequence database. The search parameters included: precursor mass tolerance 10 ppm and 0.02 Da for fragments, 2 missed trypsin cleavages, oxidation (Met) and acetylation (protein N-term) as variable modifications, carbamidomethylation (Cys) as a static modification (Trzeciecka & Bhattacharya, 2019). Protein annotation analysis was performed on GO terms using Uniprot KB and DAVID bioinformatics databases.…”
