Dasatinib reverses drug resistance by downregulating MDR1 and Survivin in Burkitt lymphoma cells

Abstract: Background: Current chemotherapies for Burkitt lymphoma (BL) have dramatically improved its clinical outcome. However, chemoresistance can lead to chemotherapy failure and very poor prognosis; thus, novel strategies are urgently required for patients with drug-resistant BL. To investigate the mechanisms underlying drug resistance in BL, we established drug-resistant BL cell lines: HS-Sultan/ADM (adriamycin-resistant), HS-Sultan/VCR (vincristineresistant), HS-Sultan/DEX (dexamethasone-resistant), and HS-Sultan/… Show more

“…Although P-gp is overexpressed both in GC- resistant and sensitive pre-B ALL cells, its activity does not correlate with GC sensitivity ( 87 , 201 ). As it was demonstrated recently, inhibition of an upregulated MDR1 in a GC-resistant B-ALL sensitized cells to DEX ( 202 ).…”

“…25) ABCB1 (member of the ATP binding box B subfamily 1) belongs to the ABC protein family and is one of the most important mechanisms for MDR. 26) In a study of tumor specimens in patients with colorectal cancer, ABCB1 and LC3 (light chain 3) were positively correlated with the expression levels of Beclin1 and Rictor. Instead, there is a negative correlation between the expression levels of ABCB1 and Raptor (regulatory-binding protein of mammalian target of rapamycin (mTOR)).…”

“…Second, the AKR1C3 single nucleotide polymorphism may influence the survival outcomes of patients receiving the CHOP regimen [ 69 ]. Finally, the role of ABC transporters in cancer responses to ANT-containing chemotherapy has been suggested in DLBCL [ 70 ], MCL [ 71 ], and Burkitt lymphoma [ 72 ]. In addition to the primary indication of BTK inhibitors in CLL and lymphomas, their efficacy has recently been demonstrated in AML and solid tumours [ 73 , 74 , 75 , 76 ].…”
