Das isoliert perfundierte Hinterbein der Ratte als Modell zur Untersuchung des Muskelstoffwechsels

Abstract: The Isolated Perfused Hindquarter of the Rat as a Model for Studying Muscle MetabolismSummary. 1. A detailed description is given of the perfusion technique of the rat hindquarter with a semisynthetie medium. 2. The concentrations of creatine phosphate, ATP and ADP in the muscle tissue after 60--180 min of perfusion were identical with those measured in vivo.3. The glucose uptake of the resting hindquarter of male fed rats was 175.2 tzMol per 30 g/hr 63% of the oxygen consumption accounted for glucose oxidatio… Show more

“…Similar results were reported with the perfused rat heart [15] and in in vivo experiments [11]. In order to investigate a direct action of buformin on glucose utilization in skeletal muscle, the isolated perfused hindlimb of the rat was used as a suitable model for metabolic studies [13,14]. In this study we report the effect of low and high doses of buformin, administered orally for one or several days, on the metabolism of the perfused resting hindlimb of normal rats.…”

“…Insulin (1 I.U.jl50 ml) stimulated the uptake 3-fold to about 5.1 pmol x min-'. The higher stimulation of glucose uptake reported by others [15,35,36] might be attributed to higher glucose concentrations in the perfusate and to the intact innervation [15,35].…”

“…It is also a most useful indicator for the finctional integrity of the perfused hindquarter [15,35]. Generally, a flow rate through the hindquarter of more than 8 ml x min-' is held necessary [15,35 -371 using an erythrocyte-containing perfusate.…”
