DARPP-32, a dopamine- and adenosine 3':5'-monophosphate-regulated phosphoprotein enriched in dopamine-innervated brain regions. III. Immunocytochemical localization

Abstract: Immunocytochemical studies have been carried out to determine the regional and cellular distribution of DARPP-32, a protein the phosphorylation of which can be regulated by dopamine and cAMP in intact cells. These immunocytochemical studies indicate tha DARPP-32 is localized primarily in those brain regions enriched in dopaminergic nerve terminals. Moreover, the staining pattern supports the conclusion that the DARPP-32 is present in dopaminoceptive neurons, i.e., neurons that receive a dopamine input, and tha… Show more

“…At 4 months, the grafts also contained a population of cells expressing the specific striatal projection neuron marker DARPP-32 not only in the striatum but also in the cortex, where only weakly stained DARPP-32þ neurons are normally seen [42]. Two important conclusions of relevance for cell replacement strategies can be drawn from our data.…”

“…DARPP-32 (dopamine-and cAMP-regulated phosphoprotein, Mr=32,000) 8 is a prominent mediator of DA signaling in the striatum 9 . DARPP-32 is highly enriched in striatal GABAergic mediumsize spiny neurons (MSN) 10 . Following activation of DA D1 receptors (D1R), DARPP-32 is phosphorylated by cAMP-dependent protein kinase (PKA) at Thr-34 and converted into a potent inhibitor of the multifunctional serine/threonine protein phosphatase-1 (PP1) 11 .…”

“…Dopamine and cAMP-regulated phosphoprotein M r 32 kDa (DARPP-32); is a neostriatum-enriched substrate for PKA (Walaas et al, 1983;Ouimet et al, 1984). Phosphorylation at Thr 34 by PKA converts DARPP-32 into an efficient inhibitor of protein phosphatase 1 (PP1) , a serine/threonine phosphatase enriched in the dendritic spines of neostriatal neurons (Ouimet et al, 1995).…”
